Objective: To study the expression and distribution of EphB4mRNA of renal cancer,corresponding adjacent renal tissue and normal renal tissue and its relationship with MMP-9mRNA expression of renal cancer, to investigate EphB4mRNA and MMP-9mRNA's possible roles in the renal carcinogenic mechanism and to provide a new method to diagnosis,therapy,and prognosis assessment。Methods: Speciments of renal cancer,corresponding adjacent renal tissue and normal renal tissue were obtained from 42 cases who had undergone surgical operations of renal cancer at the second hospitals affiliated to the Nan Chang university。All samples were confirmed by pathologic diagnosis.Expressions of EphB4mRNA,MMP-9mRNA were dectected by senmiquatitative reverse transcriptase polymerise chain reaction in 42 specimens of renal cancer,corresponding adjacent renal tissue and normal renal tissue,and further we investigated the relationships between the expression of EphB4mRNA,MMP-9mRNA and the clinicopathologicparmeters.explored the possible role of EphB4mRNA and MMP-9mRNA in proliferation,metastasis of renal cancer. The data were analysized by software SPSS13.0,a difference was considered signicance if the P value was below or equal to 0.05。Results:1 Expression of EphB4mRNA,MMP-9mRNA and the relationships to clinicopathologicaparameters.The rations of intensities of EphB4mRNA/β-actinmRNA,MMP-9mRNA/β-actinmRNA of renal cancer,corresponding adjacent renal tissue and normal renal tissue were 1.267±0.368,1.148±0.425,0.380±0.119 and1.415±0.289,1.296±0.360,0.433±0.120,respectively.There are significant differences of EphB4mRNA/β-actinmRNA,MMP-9mRNA/β-actinmRNA between renal cancer,corresponding adjacent renal tissue and normal renal tissue(Hc =71.31,P=0.000;Hc =82.41,P=0.001),EphB4mRNA and MMP-9mRNA were shown upregulated in renal cancer,corresponding adjacent renal tissue compared to paired normal tissue。Besides,high expression of EphB4mRNA and MMP-9mRNA were associated with deep tumor invasion,pTNM stage and histological differentiation(p<0.05),but not associated with age,gender ( P>0.05)。2 Correlation among expression of EphB4mRNA and MMP-9mRNA in renal carcinomaThere was a significant positive correlation between expression of EphB4mRNA and MMP-9mRNA in renal carcinoma(r=0.631,p<0.001)。Conclusions:1 EphB4mRNA and MMP-9mRNA were over expressed in renal carcinoma. EphB4mRNA and MMP-9mRNA were shown upregulated in renal cancer,corresponding adjacent renal tissue compared to paired normal tissue。2 EphB4mRNA and MMP-9mRNA expression were significantly correlated with progression of renal carcinoma,differentiation grades and depth of tumor invasion. but not associated with age,gender。3 There was a significant positive correlation between expression of EphB4mRNA and MMP-9mRNA in renal carcinoma , high expression of EphB4mRNA might be a cause of upregulating the MMP-9mRNA,and promote tumor celIs to cleavage or proliferate,and play an important role in tumorigenesis and progression。By jointly detecting of the expression of them, they might function as helpful markers in predicting the prognosis of renal cancer and directing the reasonable therapy for it。...
