Objective To investigate the expression of p21WAF1/CIP1 and TGF-β1 in retinoblastoma(RB) , and to correlate its expression with clinicopathological parameters of RB:pathological staging,histological phenotype,and infiltration depth,as well as the correlation between the expression of p21WAF1/CIP1 and TGF-β1 in the tissue of RB.Methods We used immunohistochemical method (SP) to measure the expression of p21WAF1/CIP1 and TGF-β1 in 46 paraffin imbeding specimens.Results 1. p21WAF1/CIP1 was mainly expressed in both nucleus and cytoplasm,22 in 46 RB specimens had positive expression , the positive ratio was 47.83%. Significant difference was found in the expression of p21WAF1/CIP1 in tumors with different pathological staging(P<0.05). There was a negative correlation between the expression of p21WAF1/CIP1 and pathological staging of RB(rs=-0.743). The positive ratio of p21WAF1/CIP1 was decreased with the advancement of pathological staging. The expression of p21WAF1/CIP1 was higher in differentiated type than undifferentiated type,the expression of p21WAF1/CIP1 was higher in without optical nerve infiltration than with optical nerve infiltration (P<0.05).2. TGF-β1 was mainly expressed in both cytoplasm and cytolemma,30 in 46 RB specimens had positive expression,the positive ratio was 65.22%. Significant difference was found in the expression of TGF-β1 in tumors with different pathological staging(P<0.05). There was a positive correlation between the expression of TGF-β1 and pathological staging of RB( rs=0.627). The positive ratio of TGF-β1 was increased with the advancement of pathological staging. The expression of TGF-β1 was higher in undifferentiated type than differentiated type, the expression of TGF-β1 was higher in with optical nerve infiltration than without optical nerve infiltration (P<0.05).3. There was a negative correlation between the expression of p21WAF1/CIP1 and TGF-β1 in RB( rs=-0.685).Conclusion 1. There was a significant correlation between p21WAF1/CIP1,TGF-β1 and clinicopathological parameters. With the advancement of RB, the expression of p21WAF1/CIP1 was decreasing, the expression of TGF-β1 was increasing.2. p21WAF1/CIP1 and TGF-β1 might participate in the genesis and the development of RB together,and the change of their expressions were closely correlated with the aggravation and the unfavourable prognosis of RB.They minght be the reference index for the prognosis and estimation of RB,and offer the new way for gene therapy.
