Object:To explore the association of lecithin:cholesterol acyltransferase(LCAT) gene polymorphism with the occurrence of familial aggregation of cerebral hemorrhage and lipid.Methods Lecithin:cholesterol acyltransferase gene 608C/T polymorphism was detected by polymerase chain reaction(PCR) and restriction fragment length polymorphism(RFLP) in 10 families of cerebral hemorrhage with family history(CHFH) including 30 cerebral hemorrhage patients(CHFH-P),38 first degree relatives(CHFH-Ⅰ),31 second defree relatives(CHFH-Ⅱ),27 third degree relatives(CHFH-Ⅲ), 100 patients with sporadic cerebral hemorrhage(SCH) and 100 healthy controls.oxidation enzymic method to detect HDL,LDL,TC,TG;immunoturbidim to apoA-Ⅰand apoB.Results1.The CC and CT genotype frequency of C608T in CHFH-P, CHFH-Ⅰ,CHFH-Ⅱ,CHFH-Ⅲ,SCH patients and control group were 0.933 and 0.067;0.948 and 0.052;0.903 and 0.097;0.926 and 0.074:0.93 and 0.07;0.96 and 0.04 respectively.While the T allele frequency of C608T in familial aggregation of cerebral hemorrhage group,CHFH-Ⅰ,CHFH-Ⅱ,CHFH-Ⅲ,SCH patients and control group were 0.967/0.033,0.974/0.026,0.951/0.049,0.964/0.036,0.955/0.045 and 0.963/0.037 respectively.There was no significant difference between the genotype distribution and the alleles frequencies in CHFH-P,CHFH-Ⅰ,CHFH-Ⅱ,CHFH-Ⅲ,SCH patients and control group(P>0.05).2.The serum level of high density lipoprotein cholesterol(HDL-C) in C608T genotype subgroup was significantly lower than that in C608C genotype subgroup of the same group in familial aggregation of cerebral hemorrhage group,sporadic cerebral hemorrhage group(P<0.05) However,other blood-fat index were not significantly different in both subgroups of each group.Blood-fat index weren't significantly different in two genotypes of control group(P>0.05).Conclusions1.There is no significant association of C608T polymorphism of LCAT gene with familial aggregation of cerebral hemorrhage in han population of Hunan province2.T alleles of C608T are possibly associated with the metabolism of HDL.
