Objective:To detect the expressions of STAT1 and PIAS1 in nomal cervical tissue,cervical intra-epithelial neoplasia tissue and early cervical squamous cell carcinoma tissue,so that to investigate the relationship between JAK/STAT1 signal transduction pathway and its repressor PIAS1 and the occurrence,development and clinicopathologic characters of cervical squamous cell carcinoma.Methods:Imunohistochemistry of SABC was adopted to detect the expression of STAT1 and PIAS1 in 30 cases of normal cercvix,45 cases of CIN and 53 cases of early cervical squamous cell carcinoma.Results:1,Expressions of STAT1 in different cervical tissues:In all the normal cervical tissues,CIN tissues and early cervical squamous cell carcinoma tissues,there were expressions of STAT1,and the immunohistochemical staining were located mainly in endochylema and several nucleus.In normal cervical tissue group,the expressions of STAT1 in HPV positive cases were significantly stronger than those in HPV negative cases(P<0.05 ).With the development of cervical diease,in these three groups,the expressions of STAT1 became weaker and weaker,and the differences between every twogroups had statistical significance(P<0.01).In CIN group,as for the expression of STAT1, CINI group was significantly weaker than normal cervicx group(P<0.05), significantly stronger than CINâ…¢and early cervical squamous cell carcinoma group(P<0.05) and compared with CINâ…¡,it is also stronger but the difference had no statistical significance(P>0.05);CINâ…¡group was significantly weaker than normal cervix group(P<0.05),significantly stronger than CINâ…¢group(P<0.05) and early cervical squamous cell carcinoma group(P<0.05);CINâ…¢group was significantly weaker than normal cervix group(P<0.05) and significantly stronger than early cervical squamous cell carcinoma group(P<0.01).And in early cervical squamous cell carcinoma group,the expression of STAT1 was related to tumor FIGO staging,pathological grade and lymph node metastasis (P<0.05). 2,Expression of PIAS1 in different cervical tissues:In all normal cervical tissues,CIN tissues and early cervical squamous cell carcinoma tissues,there were expressions of PIAS1,and the immunohistochemical staining were also located mainly in endochylema and several nucleus. With the development of cervical diease,in these three groups,the expressions of PIAS1 became stronger and stronger,and the differences between every two groups had statistical significance(P<0.01) In normal cervix group,the expression of PIAS1 in HPV positive cases were significantly stronger than those in HPV negative cases(P<0.05).In CIN group,as for the expression of PIAS1,CINâ… group was significantly weaker than CINâ…¢group(P<0.05) and early cervical squamous cell carcinoma group(P<0.05),and stronger than normal cervix group, weaker than CINâ…¡group but these two differences had no statistical significantly(P>0.05);CINâ…¡group was significantly weaker than early cervical squamous cell carcinoma group(P<0.05),and stronger than normal cervix group,weaker than CINâ…¢group,but these two differences also had no statistical significance(P>0.05);CINâ…¢group was significantly stronger than normal cervix group(P<0.01),and weaker than early cervical squamous cell carcinoma group(P<0.05).And in early cervical squamous cell carcinoma group,the expression of PIAS1 was also related to tumor FIGO staging,pathological grade and lymph node metastasis(P>0.05).3,The relationship between expression of STAT1 and PIAS1:The expression of STAT1 is negative correlated to PIAS1,coefficient of correlation-0.753,P<0.01.Conclusions:1.HPV infection in gential tract may lead to the abnormal strong expression of STAT1 and its inhibitor PIAS1.2.In normal cervical tissue,CIN tissue and early cervical squamous cell carcinoma tissue the expression of STAT1 become weaker and weaker and it is related to tumor FIGO staging,pathological grade and lymph node metastasis,so it is presumed that the abnormal weak expression of STAT1 may lead to the occurrence and development of cervical squamous cell carcinoma. 3.In normal cervical tissue,CIN tissue and early cervical squamous cell carcinoma tissue the expression of PIAS1 become stronger and stronger and it is related to tumor FIGO staging,pathological grade and lymph node metastasis,so it is presumed that the abnormal strong expression of PIAS1 may be another factor of the occurrence and development of cervical squamous cell carcinoma.
