| [Objective]To assess the most suitable way of processing blood for measuring VEGF as a marker of tumor-angiogenesis through testing and contrasting VEGF levels in plas-ma,serum or P-APR of breast cancer patients. so as to provid molecular biologic markers for early diagnosis of breast carcinoma and for estimate of prognosis diagno-sis and differential diagnosis,[Methods]Peripheral venous blood samples were collected from 42 patients with breast cancer and 20 healthy volunteers.Blood counts were performed in an automated he-mocytometer,and then the specimen was prepared into serum plasama and P-APR samples.VEGF concentrations were measured with a quantitative sandwich en-zyme(R&D Systems Minneapolis USA),a semiquantitative measure of VEGF was calculated by using microplate–reader.[Results]1. The comparison of serum VEGF, plasma VEGFand P-APR VEGF: The serum VEGF concentrations from the localized breast cancer goup did not differ signifi-cantly from those of healthy control group(P=0.054).but the difference of plasma and P-APR VEGF between both groups was statistically signicant (P=0.031,P=0.007,respectively) P– APR VEGF is more sensitive to others between the localized breast cancer group and the health control group,Both serum and plas-ma VEGF were higer in metastatic breast cancer goup than in normal con-trols(P=0.035,P=0.018, respecttively).The difference of VEGF in metastatic breast cancer goup was larger than than in healthy controls(P=0.001).2.The corelevance of the levels of VEGF and platelet or leucocyte: A correlation between serum VEGF levels and platelet counts was found in the health control group(r=0.632,p=0.003) and in the metastatic breast cancer group (r=0.547, p=0.012).and there also was a significant correlation between serum VEGF levels and leucocyte in both group(r=0.587, P =0.006;r=0.576, P=0.008).3.The corelevance of the levels of VEGF and Hemoglobin:Hemoglobin contents did not correlate with serum VEGF concetations between the health control group and metastatic breast cancer gtoup(r=0.212,P=0.370;r=0.204,P=0.389)4.The platelets from the metastatic breast cancer goup did not differ significantly from those of healthy control group(t=0.479,P=0.635).but the difference of theoretical platelet-derived VEGF between both groups was statistically significant(t=-2.115, P=0.041).[Conclusions]1. SerumVEGF content is larger in peripheral venous blood than plasma VEGF and P-APR VEGF,so it has good sensitivity and could be more easily tested,it could be serve as a screening program to prevent or detect breast cancer,Compare with se-rum plasma, P-APR is the best section assaying VEGF.in peripheral blood.2. The level of VEGF in peripheral venous blood positively correlates to platelets and leucocytes,it increased with increasing platelet or leucocyte counts.3. In peripheral venous blood,VEGF exists in platelets and leucocytes possibly, but we don't know if platelets secrete VEGF itself or just as transpoter of VEGF.This study had shown that platelets may be scavenger of VEGF on tumor growth.4. .We found that Hemoglobin contents did not correlate with serum VEGF conceta-tions in this study.5. The specimens in this research were collected before operation,so the conclusion has less value to prognosis relatively. The number of cases were small, as for ex-ploratory research, its conclusions still need more cases and prospective experi-mental studies to confirm. |
