Pathophysiology of cerebral infarction involves a number of aspects, one of which is the immune system activation after cerebral infarction, resulting in non-specific inflammatory response. This pathological process is considered to be one key step in acute ischemic brain injury and it is receiving increasing attention. At present, mild hypothermia is used as a clinical treatment in department of neurology. Both at home and abroad,the role and mechanism of mild hypothermia for its treatment to acute cerebral ischemic injury has been researched in depth. That mild hypothermia has obvious protective effect on the acute cerebral ischemic injury has been confirmed, and that this protective effect may be related to inhibiting adhesion and aggregation of inflammatory cells in ischemic vein, subsequent in ischemic brain parenchymal, especially in ischemic surrounding area. However, effect of mild hypothermia on adhesion between leukocytes and vascular endothelial cells has been researched rarely. In this paper, the influence of mild hypothermia on adhesion between hypoxia/reoxygenation endothelial cells and neutrophil is observed, effect of mild hypothermia on serum soluble intercellular adhesion molecule-1 and L-selectin in patients with cerebral infarction is observed. Then discuss the influence of mild hypothermia on inflammatory injury after cerebral infarction.Objective:1. Explore the effect of mild hypothermia to the expression of adhesion molecule-1 on hypoxia/reoxygenation endothelial cell.2. Explore the effect of mild hypothermia to adhesion between hypoxia/reoxygenation endothelial cell and neutrophil.3. Explore the effect of mild hypothermia on cerebral infarction in patients with serum soluble intercellular adhesion molecule-1 and L-selectin levels.Research methods:1. Using ECV304 Endothelial Cells as researchable object, utilizing simple and anoxic box to build the model of hypoxia / reoxygenation endothelial cell, appliing mild hypothermia intervene hypoxia / reoxygenation endothelial cell.2. Appling western blot determine the ICAM-1 expression of hypoxia / reoxygenation endothelial cells, appling cell counting method evaluate adhesion rate between endothelial cells and neutrophil.3. Appling brain cooling device do local mild hypothermia therapy in patients with cerebral infarction.4. Using ELISA method, the serum contents of ICAM-1 and L- selectin on patients with acute cerebral infarction were measured at 1,3 ,5 and 7 days after infarction.Study Results:1. Mild hypothermia can reduce the expression of ICAM-1 on hypoxia / reoxygenation endothelial cells and effectively inhibit the adhesion between hypoxia / reoxygenation endothelial cells and neutrophil. 2. Mild hypothermia can inhibit the expression of ICAM-1 and L-selectin in patients with cerebral infarction.Conclusion of the study:Mild hypothermia can reduce the inflammatory response after cerebral infarction, and its mechanism may be that mild hypothermia can reduce the adhesion between vascular endothelial cells and neutrophil in cerebral infarction zone and inhibit expression of adhesion molecule ICAM-1 and L-selectin.
