Objective:To investigate the changes of AR,O2-·,SOD,CAT activity in diabetic microangiopathy.O2-·is a oxidative stress product,SOD and CAT are oxidizing material catabolic enzymes.To interpret exceptional biochemistry change antioxidation in vessels microenvironment.Identify the relation between the changes of AR activity and hyperglycosemia,oxidative stress.Methods:Healthy adult male 120 SD rats were randomly divided into normal control group and diabetic group.The diabetic group feed with high glucose and high fat bait vessel,diabetic rats were induced by injection of streptozotocin(STZ) intraperitoneally.The ones whose blood glucose is above 13.8 is succeed.Select 15 SD rats(blood glucose:13.8~16.7) is diabetes group Low.Select 15 SD rats(blood glucose>16.7) is diabetes group High.Then take blood,The activity of AR in the blood and brain tissue were measured by NADPH decreasing test method.The activity of SOD and CAT were measured by bioehemieal test method in eaeh group of the experiment.Results:①Diabetic SD rat group L the activity of O2-·(95.874±24.58) is higher than control group(65.944±11.38) significantly(P<0.05),Diabetic SD rat groupH the activity of O2-·(105.434±22.86) is higher than control group significantly(P<0.01), the difference with the diabetic SD rat group L did not achieve statistical significence(P<0.05).②Diabetic SD rat group L the activity of SOD(23.65±7.62) is lower than control group(38.93±12.01) significantly(P<0.05),Diabetic SD rat group H the activity of SOD(16.41±6.82) is lower than control group significantly(P<0.05),the difference with the diabetic SD rat group L achieve statistical significence.(P<0.05)③Diabetic SD rat group Low the activity of serum AR(4.16±0.96U/ml) is higher than control group(2.44±1.27U/ml) significantly(P<0.05),Diabetic SD rat group High the activity of AR(4.44±1.05U/ml) is hinger than control group significantly (P<0.05).④the difference of serum CAT activity in every group did not achieve statistical significence(P>0.05)Conelusion:①the activity of O2-·in diabetes group Low and High is higher than normal control group,the diabetes group High is more significant,indicate when in diabetic microangiopathy,oxidative stress strengthen,but the activity do not change with blood glucose concentration.②the activity of SOD in diabetes group Low and High is lower than normal control group,indicate when diabetic microangiopathy,hyperglycosemia is main reason of ROS priming,the activity of SOD is different significantly between diabetes group low and high,the SOD activity was associated with blood glucose concentration.③the activity of AR in diabetes group is higher than control significantly,but this difference can not be observed between the diabetic group Low and diabetes group High.indecate when diabetic microangiopathy,AR change with oxadation and oxadation resisting system,diabetic microangiopathy and nervous tissue damage is correlated close with O2-,SOD,AR,SOD is most significantly,it is maybe the key reason in disbalance resulting complication occurred,the difference between the diabetic group Low and diabetes group High indicate AR activity do not change with blood glucose concentration.The rate of increase of AR activity in brain tissue is higher than in serum,indicate AR is correlated close with diabetic microangiopathy and nervous tissue damage.④the difference of serum CAT activity in every group did not achieve statistical significence,maybe there is lab error. Objective:To investigate the changes of AR and oxidative stress product O2-,H2O2,CAT,GSH activity in type 2 diabetes patients with cerebral vascular accident..To interpret exceptional biochemistry change antioxidation in Cerebral vessels microenvironment and identify the relation between the changes of AR activity and hyperglycosemia,oxidative stress.Methods:240 cases of type 2 diabetes patients were selected from Geriatrics in the Second Affiliated Hospital of Kunming Medical College,(December,2008 -May,2009),according to the Complication of diabetes,4 group were divided including to the control group,each group with 60 cases,randomly selected men and women,aged 40-80 years old.Diagnosis of diabetes mellitus is based on ADA diagnostic criteria,microvascular complications are based on clinical symptoms and laboratory test results,CT and MRI were checked in diabetes with cerebral vascular accident patients to do for brain tissue morphology,all cases have records of Age,sex, duration of diabetes,body mass index(BMI),fasting blood glucose,triglycerides, HDL,LDL,diabetic plus measuring glycated hemoglobin and glucose,Ketone.All groups were excluded autoimmune diseases and inflammatory diseases,the control group selected for the normal physical examination to exclude cardiovascular, diabetes and other diseases.O2-,H2O2,CAT,GSH activity and AR were checked in all cases.Results:1,General:Among the four groups,age,sex,body mass index,FBG have no significant differences(P>0.05);For serum triglyceride and low-density lipoprotein levels,the diabetes groups were higher than the control group,but there were no statistically significants,(P>0,05),and no significant differences among the three diabets groups(P>0.05);For serum high-density lipoprotein levels,the diabetes groups were lower than the control group,but there were no statistically significants, (P>0.05),and no significant differences among the three diabets groups(P>0.05).2,Comparison of aldose reductase activity in each group:three groups in diabetes were higher than the normal control group,there were significant differences (P<0.05);DC group and DS group were higher than DM group,with significant differences(P<0.05),but there was no significant difference between DC group and DS group(P>0.05).3,Comparison of O2-activity in all groups:three groups in diabetes were higher than the normal control group,there were significant differences(P<0.05);DC group and DS group were higher than DM group,with significant differences(P<0.05),but there was no significant difference between DC group and DS group(P>0.05).4,Comparison of H2O2 concentration in each group:three groups in diabetes were higher than the normal control group,there were significant differences(P<0.05);DC group and DS group were higher than DM group,with significant differences(P<0.05),but there was no significant difference between DC group and DS group(P>0,05).5,Comparison of CAT activity in each group:three groups in diabetes were lower than the normal control group,there were significant differences(P<0.05);DC group and DS group were lower than DM group,with significant differences(P<0.05),but there was no significant difference between DC group and DS group(P>0.05).6,GSH concentration in each group comparison:three groups in diabetes were lower than the normal control group,there were significant differences(P<0.05);DC group and DS group were lower than DM group,with significant differences(P<0.05),but there was significant difference between DC group and DS group(P<0.05).Conelusion①For H2O2,O2-,the oxidative stress product,three diabetes groups in diabetes were higher than the normal control group,DC group and DS group were higher than.DM group,Shows that there is diabetic microangiopathy,leading to increased oxidative stress,O2-activity increased in the event of brain injury,the stronger the role of oxidative stress,free radicals generated more and more harmful to the body.This change is more obvious when complicates with microvascular disease and nerve tissue damage.②For activity of GSH concentration,three diabetes groups was significantly lower than the control group,DS group was the lowest.It indicated that in high glucose environment,oxidative stress is strengthened,with oxidation and antioxidant balance broken,ROS increased,antioxidants,consumpted,for lowest levels when happened chronic complications in diabetes mellitus syndrome③For activity of CAT concentration,DC and DS group were lower than CO group,but DM group had no significant difference with CO group,when oxidative stress occured,with the duration of the extension and complications,antioxidation become less,these changes are more obvious when capillaries and nerve tissue damaged,also confirmed the relationship of oxidative stress and diabetes complications.④For AR activity,three diabetes groups in diabetes were higher than the normal control group,DC group and DS group were higher than DM group,It indicated that with diabetic microangiopathy and nerve tissue injury happened,oxidation and antioxidant system changes,AR also change.Diabetic microangiopathy and related organizations,lesions and O2-,H2O2,CAT,GSH,AR oxidative stress is closely related to the material.Cerebral vascular accident group the lowest AR activity, indicating that AR and diabetic microangiopathy and nerve tissue injury is closely related to,and with the duration of the extension and anti-oxidation gradually decreased,AR activity was decreased.⑤When oxidative stress happende,free radicals increased,antioxidative capacity declined,oxidative and antioxidant balance was broken,leading to diabetic microvascular complications and the incidence of stroke,oxidative stress plays an important role in diabetic complications.
