Objective To explore the change of oxidative stress in subjects with type 2 diabetes in early stage and the modification of Six Drugs Containing Rehmanniae Preparata(SDCRP) and Tablet of ginkgo Leaf after 18 month treatment.Methods The study was an 18-month double-blind, randomized, parallel controlled add-on trial. A total of 103 subjects with diagnosed in type 2 diabetes with previous treatment of hypoglycemic agents in early stage were recruited, whose differentiation is deficiency of kidney yin type, accompanied by internal heat and stagnation. They were randomized into two groups, one group receiving SDCRP and Tablet of ginkgo Leaf while the other group receiving placebo. 87 subjects completed the trial, 43 received SDCRP and Tablet of ginkgo Leaf and 44 received placebo. At twelfth month, SOD, MDA were measured, and ①Glucose Metabolism: FBG, PBG, HbA1C; ②Lipid Metabolism: TC, TG, FFA; ③Inflammation Factors: TNF- α , IL-1 0 ; ④ Carotid intinal medial thickness(IMT)were also measured. After 6 month follow-up, the above profiles were repeated after cessation of medication.Results ①There were significant increase in SOD and MDA (P<0.01). ②Stratified with TG of twelfth month, when TG>1.7mmol/L, MDA increased greatly in trial group, while reduced in control group. ③There were increase in FBG in both group. PBG increased significantly in trial group (P<0.05) . But it was shown no significant difference in HbA1C between twelfth and eighteenth month. ④LDL-C increased greatly in trial group, while slightly in control. ⑤ It was shown difference between twelfth and eighteenth month of the induction of IL-1β in Both group and TNF- α in trial group. There were significant increase in IMT (P< 0.01).Conclusions ①Oxidative stress increases in type 2 diabetes in early stage. ②It is showed a tendency that SDCRP and Tablet of ginkgo Leaf can retard the progress of lipid peroxidation in diabetes with hypertriglyceridemia.③It is also showed a tendency that the two traditional medicine can retard the increase of TG, LDL-C and the stegnosis of vessel, and lower the level of inflammatory. ④ The two traditional medicine improve the condition of oxidative stress maybe through lowing hyperglycaemia and FFA.
