Objective:(1) To establish a reliable rats model of orthotopic liver transplantation with non-heart beating donors.(2) In this model,to compare the effects of University of Wisconsin(UW) solution and self-designed KYL solution on preservation of livers from non-heart-beating rats.Methods:The model was established with modified double-cuff method.Advance by the dorsal penile vein donor input 1ml normal saline containing heparin(including heparin 1250U/ml) to the donor of systemic heparin,The thoracic descending aorta and the bottom of rat heart were cross-clamped to induce warm ischemia as a NHBD model.In this study Wistar rats were randomly assigned to warm ischemia time points two large groups,warm ischemia time was 10min(R10),30min(R30).After flushed with the UW and self-designed KYL cold preservation solution,NHBD liver were stored at 4℃for 1h,3h or 6h.Then they were transplanted.Weight changes,liver enzymes(AST and ALT),ET-1 level in serum at 24h,3d time point after liver transplantation were measured.Cytokines TNF-a and IL-1 level in the serum were measured with ELISA method.Pathological examination of HE staining and Survival in rats were measured too.Comparison of various aspects of self-designed preservation solution KYL solution and UW solution on NHBD liver graft preservation. ResultS:(1) We established NHBD model,1 week survival rate of 80%。(2) under the conditions of warm ischemia 10min,respectively,use KYL solution and UW solution to cold preservation of the NHBD 1h,3h,6h,after the 24h,3d from the two time points to examine the level of liver enzymes(AST,ALT),ET-1 levels,TNF-a levels,IL-1 levels and short-term survival after surgery were not significantly different(P>0.05).there are no difference between UW solution and KYL solution in 10min under warm ischemia time,cold preservation NHBD1h, 3h,6h effect.(3) under the conditions of warm ischemia time 30min,respectively,use KYL solution with UW solution to cold preservation of the NHBD 1h,after surgery,at the 24h,3d from the two time points to examine the level of liver enzymes(AST, ALT),ET-1 level,TNF-a levels,IL-1 levels were not significantly different(P>0.05).there are no difference between UW solution and KYL solution in 10min under warm ischemia time,cold preservation NHBD1h effect.In cold preservation 3h,6h group were significantly different between groups(P<0.05).Decreased significantly after short-term survival.In each group in cold preservation 3h,6h, UW solution was better than KYL solution.Conclusions:(1) The model which We established NHBD donor liver transplantation is a good model for NHBD donor liver transplantation for basic research.(2) In 10min of warm ischemia time conditions,the use of KYL solution can preservation the NHBD rat liver tolerance to cold preservation time 6h.(3) under the conditions 30min of warm ischemia with KYL solution preserved NHBD rat liver 1h,the effect is satisfactory.Cold preservation time 3h,the effect began to significantly deteriorate.Cold preservation time 6h effect of cold preservation of NHBD worse.Therefore,the proposed use of KYL solution in warm ischemia time was 30min save more than NHBD liver,the cold preservation time of 1h,maximum is 3h.(4) Since this study is based on the basis of animal experiments,and the sample size too few,in fact,the value to be used for further experimental and clinical validation.
