The dry buds of Lonicera macranthoides Hand.-Mazz., a resource of "Shan Yinhua" recorded in volume I of Pharmacopoeia of People's Republic of China (2005), has been used as the mainstream species of "JinYinhua" in the south and southwest of China. The medicinal materials has been used widely in China for thousands of years for the effects of bacteriostasis, anti-virus, anti-inflammation, antioxidant and hematischesis.In order to control the quality of L. macranthoides, analytical methods were established for the fingerprint of L. macranthoides and simultaneously determining the contents of chlorogenic acid and galuteolin in L. macranthoides.The contents of chlorogenic acid and galuteolin were mensurated in different batches of L. macranthoides by HPLC. The results showed that samples of L. macranthoides from different habitats were greatly different in the contents of chlorogenic acid and galuteolin, and that medicinal materials should be collected in the green bud time at first stubble of plants of 3 years old and processed with kill-enzyme torrefaction immediately. In this way, medicines could be obtained not only containing more active ingredients such as chlorogenic acid and galuteolin, but also with a bright color.The method of fingerprint analysis of L. macranthoides by HPLC has been estabished. The HPLC conditions were optimized as follows:at on a Kromasil C18 column (4.6 mm×250 mm,5μm), acetonitrile and 0.1% phosphoric acid solution as mobile phases in gradient mode, the flow rate being 1.0 mL-min-1 and the column temperature being 30℃. The gradient program of eluent was 0-10 min,89%; 10-25 min, 89%-80%; 25-38 min,80%-77%; 38-57 min,77%-71%; 57-60 min,71%-89%. The detection wavelength was set at 238 nm.30 batches of L. macranthoides from Xiushan and 13 batches from different habitats in other regions of China were analyzed with the HPLC fingerprint method. With the reference of chlorogenic acid, the similarity of all above chromatograms were compared and the fingerprints of L. macranthoides were built with 14 common peaks.Combining comparison of the fingerprints of 5 species included in the materials of "ShanYinhua" and "JinYinhua" recorded in volume I of Pharmacopoeia of People's Republic of China (2005 edition) with analysis of the quantitative determination results of chlorogenic acid and galuteolin, it was concluded that L. macranthoides should be differentiated from "Jin Yinhua" or "Shan Yinhua". So, we suggest that L. macranthoides should be separated from ShanYinhua and used as a stand-alone medicine in next edition of Pharmacopoeia of People's Republic of China.Both the method of the content detecting and the HPLC fingerprint establised had satisfactory precision.stability and repeatability. It can be used to control the quality of L. macranthoides and differentiate positive or negative herbs in order to ensure efficacy and safely use in clinic.
