This thesis mainly deals with two Traditional Chinese Medicines, Taraxacum mongolicum Hand-Mazz (Compositae) and Isatis indigotica Fort. (Cruciferae). The thesis is composed of two chapters.Part ⅠThe Chemical Constituents of Taraxacum Mongolicum Hand-Mazz.Herba Taraxaci, which has a long history as a Traditional Chinese Medicine, has different species as well as rich resources in China. Among which, the most popular species is Taraxacum mongolicum Hand-Mazz. Chemical investigations revealed that it contains triterpenoids, flavonoids, coumarins, phenolic acids, sesquiterpene lactones, plant sterols, long-chain fatty acids and cartenoids. Pharmacological studies showed its activities of inhibiting bacteria, anti-inflammation, resisting endotoxin, strengthening stomach, normalizing the secretion and discharge of bile, improving lactation, inhibiting tumors and immunological regulation. Clinically, it is frequently used to treat upper respiratory infection, pharyngo-laryngitis chronica, acute and chronic gastritis, mastitis, hepatitis, cholecystitis and urinary tract infection. In order to find out the active components, we studied and optimized the extracting and refining methods according to pharmacological experimental results. After obtaining the active part, the chemical investigation work including purification and analysis were carried out. The main research results are as below:1. Finding out the optimized extracting and refining methods;2. 15 compounds have been isolated from the active part, and 11 structures were identified, by chemical and spectral methods, as taraxasteryl acetate (1), Ψ-taraxasteryl acetate (2), caffeic acid (3), artemetin (4), luteolin (5),luteolin-7-O-P-D-glucopyranoside (6), esculetin (7), quercetin (8), daucosterol (9), /?-sitosterol (10), stigmasterol(ll). Other two compounds were characterized by comparing to the standard samples with HPLC as ferulic acid (12) and chlorogenic acid (13).3. The contents of total phenolic acids and flavonoids were measured by the means of UV spectrophotometry using caffeic acid and rutin as control standard, and the contents to be 36.27% was 49.90%, respectively.4. The contents of caffeic acid and luteolin-7-O-P-D-glucopyranoside were measured by HPLC chromatography with external standard. The absolute contents are 0.6% and 1.0% respectively.Part IIStudies on HPLC Fingerprint of Isatis Indigotica Fort.Radix Isatidis is the dry root of Isatis indigotica Fort.( Cruciferae). It is one of the most common used Traditional Chinese Medicine with the effect of clearing away heat, detoxicating, cooling blood and anti-inflammation. In clinical, it is widely used to treat various infectious deseases such as influenza, hepatitis, parotitis, epidemic meningitis, tonsillitis.As a common medical material, there are a lot of Radix Isatidis available in market, but their quantities are usually different. Phytochemical investigations revealed that there exist abundant indole and quinoline alkaloids in its low polarity part and they are the representative secondary metabolites for Isatis indigotica Fort. Up to now, the quantities control method of TCM preparations that contains Radix Isatidis is still limited to measure the content of indirubin or indigotin. In order to find out the commonness^f£) and difference of Radix Isatidis medical material circulating in the market, we collected 11 batches materials from different provinces of China, studied its HPLC fingerprintchromatography and found out 12 common peaks containing indirubin. The research results:1. The established HPLC fingerprint chromatography can be used to identify the Radix Isatidis and reflect the quantitiy.2. The constituent sorts of Radix Isatidis circulating in the TCM markets were similar, but the contents were quite different.
