Objective: Ixeris chinensis?Thunb.? Nakai and Sonchus brachyotus DC. belongs to the Compositae plants, had been used as North Patrinia herb. The working is on the research of chemical constituents and pharmacological action in common, for advising the basis for building standard of the two herbs. Methods: Using the method of microscopic identification to identificat the internal structure of Ixeris chinensis?Thunb.? Nakai and Sonchus brachyotus DC., observed the common points and differences between Ixeris chinensis?Thunb.? Nakai and Sonchus brachyotus DC.; optimization of extraction methods, using LC-MS to detect the extract of Ixeris chinensis?Thunb.? Nakai and Sonchus brachyotus DC., analyzing the common chemical composition; building the HPLC-fingerprint of Ixeris chinensis?Thunb.? Nakai and Sonchus brachyotus DC.; comparative analysis and pharmacological effects of extract of Ixeris chinensis?Thunb.? Nakai and Sonchus brachyotus DC. through model of liver injury in mice induced by carbon tetrachloride; establishment of xylene induced mouse ear edema model and carrageenan induced rat paw swelling model of acetic acid and acetic acid induced writhing test, increased capillary permeability in mice caused by experiments, analysis the anti-inflammatory pharmacological action of Ixeris chinensis?Thunb.? Nakai and Sonchus brachyotus DC.. Results: through optical microscope can be observed to Ixeris chinensis?Thunb.? Nakai and Sonchus brachyotus DC. internal structure have some similarities, such as vascular bundles are collateral, stomata are inequalities accounted for the majority, catheters have threaded conduit accounted for the majority, that is, they all contain inulin and non glandular hairs, but there are subtle differences, such as epidermal cell anticlinal walls of bending degree, Sonchus brachyotus DC. slightly bent and Sonchus brachyotus DC. cells containing a lot of transparent particles, and the number of vascular bundle of different, Ixeris chinensis?Thunb.? Nakai vascular bundles 3, Sonchus brachyotus DC. have 3-4 beam.UHPLC-Q Exactive Orbitrap-HRMS detection results: Ixeris chinensis?Thunb.? Nakai and Sonchus brachyotus DC. in the same chemical compositions, they are chlorogenic acid, cichoric acid, galuteolin, isochlorogenic acid C and Apigenin-7-O-?-D-glucuronide. Cichoric acid and isochlorogenic acid C was first discovered in Chinese Ixeridium and Sonchus brachyotus DC..Ixeris chinensis and Sonchus brachyotus DC. characteristic peak is established through the analysis of 10 batches of the bitter endive and chicory HPLC chromatogram, the Ixeris chinensis got 10 characteristic peaks, Sonchus got 8. Ixeris chinensis and Sonchus brachyotus DC.. have 4 common characteristic peaks, corresponding chlorogenic acid, cichoric acid, luteolin, isochlorogenic acid C.HPLC method for the simultaneous determination of Ixeris chinensis and Sonchus brachyotus DC. chlorogenic acid and cichoric acid and galuteolin, isochlorogenic acid C content, column chromatography for waters C18?3.5 m, 4.6 x 150mm?, flow phase A for methanol and B is 0.5% acetic acid aqueous solution, gradient elution?015min, 10%25%A; 1535min, 25%35%A; 35min50min, 35%50%A; 50min55min, 50%70%A?. Detection wavelength at 330 nm. The flow rate was 1.0m L·min-1; column temperature 30 ?. The results of the method show that the precision of the instrument is good, the repeatability, stability and recovery rate of the test method are all within the specified range. Four chemical components in Ixeris chinensis: chlorogenic acid 6.087 mg·g-1, cichoric acid 9.646 mg·g-1, galuteolin 6.328 mg·g-1, isochlorogenic acid C 0.662 mg·g-1; in Sonchus brachyotus DC. 4 chemical composition content: chlorogenic acid 3.555 mg · g-1, cichoric acid 15.068 mg · g-1 mg-l g-1, galuteolin 6.251 mg · g-1, isochlorogenic acid C 0.580 mg·g-1. Different harvesting time Ixeris chinensis and Sonchus brachyotus DC. four chemical composition content is also different, in samples harvested in July, the contents of these four components reached highest in Ixeris chinensis in July, August and September harvest samples measured content reaches a plateau, in Sonchus brachyotus DC. is a downward trend.Liver in pharmacological experiments, Ixeris chinensis and Sonchus brachyotus DC. high, low dose group and model group were significant differences?P?0.01?; Sonchus brachyotus DC. high dose group(1.95 g·kg-1) is better than that of Ixeris chinensis high dose group?P?0.05?; low dose(0.78 g·kg-1) when administered two and no significant difference?P?0.05?. Pharmacological experiments: xylene induced mouse ear swelling model of chicory extract high dose group was better than that of Ixeris chinensis extract high dose group?P ?0.05?; rat paw edema model induced by the carrageenan, the bitter indica high, low dose group had no significant difference?P ?0.05?; acetic acid induced peritoneal capillary permeability of mice increased model, chicory extract high dose group was better than that of Ixeris chinensis high dose group; acetic acid induced writhing model, Sonchus brachyotus effect of high dose group was better than that of Ixeris chinensis extract high dose group. Comprehensive analysis of pharmacological experimental data show that Ixeris chinensis and chicory has good protective and anti-inflammatory effects, and Sonchus brachyotus DC. hepatoprotective, anti-inflammatory effect in a certain extent better than the Ixeris chinensis Conclusion: this experiment from the aspects of chemical constituents and pharmacological effects compared Ixeris chinensis and Sonchus brachyotus DC. similarities and differences were established HPLC fingerprint and using for determination of chemical components, and provides a basis for the future development of Ixeris chinensis and Sonchus brachyotus DC. common quality standards and new drug development.
