[Objective]Latent autoimmune diabetes in adults (LADA) is one of the subtypes of type 1 Diabetes, which is defined as initially non-insulin requiring diabetes diagnosed in people aged 30-50 years with antibodies. It is insulin-independent at the first, and then the patients' islet beta cells are impaired faster than the type 2 diabetes patients'. Ever since CD4+CD25+regulatory T cells (Treg) were found in autoimmune system and it can induce auto-or nonauto-immune tolerance. The characteristics and functions of Treg are closely related to surface molecules and cytokines, such as IL-4, IL-10 and TGF-beta,etc.According to recent researches, Treg is also correlated with autoimmune diabetes. Our researches aim to investigate changes and significance of IL-10 and TGF-βin peripheral blood of latent autoimmune diabetes in adults (LADA) patients.[Methods]Following certain standers and collecting peripheral blood sample of 30 LADA patients,30 type 2 diabetes and 30 controls from the endocrinopathic ward and clinic of Shandong provincial hospital and the medical examination center of Shandong provincial hospital from January 2008 to January 2009. The mean age and course of disease in three groups have no significant difference. To measure fasting blood glucose,2h blood glucose, fasting insulin,2h insulin, fasting C peptide,2h C peptide, GAD, ICA and IAA, all samples were delivered to the department of radioimmunity. The serum IL-10,TGF-beta levels were measured by ELISA kits which were purchased from R&D company, USA. The HOMA formula was used to calculate the insulin releasing index (HOMA-IS) in order to evaluate the function of islet beta cells.[Results]IL-10 in the LADA group was lower than that in the T2DM group but was higher than that in the control group. TGF-βhad no significant difference between the LADA group and the T2DM group. IL-10 had a positive correlation with LnHOMA-IS(r=0.314, P=0.017) in LADA group.[Conclusion]①The result which IL-10 in the LADA group was lower than that in the T2DM group but was higher than that in the control group suggests that an imbalance of cell immune exists in LADA patients, which may lead to the impaired autoimmune tolerance.②The result that IL-10 had a positive correlation with LnHOMA-IS suggests that IL-10 may promote the generation and development of LADA, which is a significant reason for the impaired islet beta cell functions in LADA patients.
