Objective: This study is to investigate the hyperbaric oxygen's protective effects on the function of neuromotor and apoptosis of neural cell, when the model established had been cured at different time. Then the mechanism underlying the treatment of HBO to spinal cord injury was explored.Methods: 1, 55 adult male Sprague-Dawley rats weighting about 300±10g were randomly divided into 11 groups: The normal control gro- up(n=5); the SCI group (n=25), 1st day, 4th day, 7th day, 10th day, 14th day groups; and the SCI treated with HBO group (n=25), 1st day, 4th day, 7th day, 10th day, 14th day groups;. 2, the model of Spinal cord injury was established according to the improved Allen method. The nerve injury was evaluated via the Basso-Beattie-Bresnahan (BBB) scoring system and Rivlin tiltboard experiment. Apoptosis of the injured spinal cord tissue cell was examined by the terminal deoxynucleotidal transferase-mediated Dutp-biotin nick end abeling (TUNEL) reaction. The expression of caspase-3 was determined by immunohistochemical analysis. The data were analyzed by SPSS statistics software.Result: 1, Compared to SCI group,spinal cord injury by hyperbaric oxygen therapy,nerve motor function are improved.The angels of Rivlin,s tiltboard experiment and the scores of BBB are all significantly increased(p<0.05).2,Compared to the control group, the number of apoptotic cells and expression of the caspase-3 were increased in the SCI groups. TUNEL-positive apoptotic cells were detected in SCI group of the 1st day. The apoptotic cells in SCI group 4th day increased, and then gradually decreased; there is still a small amount of apoptotic cells in SCI group 14th day. 3, compare to the non-HSO treated group, the number of apoptotic cells and the expression of caspase-3 were significantly decreased. 4, the significant difference of Apoptotic cells and caspase-3 mRNA expression were detected between the HBO treated group and HBO untreated groups.Conclusions: 1, Analysis of the result indicated that hyperbaric oxygen therapy on spinal cord injury could significantly lessen the lesion of neuromotor function.2, Neuronal apoptosis is the major way of the secondary cell death after spinal cord injury.3, apoptosisof the rats'neurons would be inhibted by Hyperbaric oxygen after the spinal cord injury, thereby, the secondary damages of nerve cells could be reduced. 4, the pretective effect of hyperbaric oxygen on the neurons might be achieved by inhibiting the expression of caspase-3 mRNA and its induced apoptosis.
