The first part: Rat C6 glioma growth law of dynamic MRI imaging observations and pathologyObjective: Establish a stable, reliable and C6 rat brain glioma model, and MRI with the clinical observation of the growth of glioma tumor variation, in order to further human gliomas in vivo provide the basis for experimental study.Methods: First, in cultured rat C6 glioma cells, and then collected at the logarithmic phase of C6 glioma cells which were washed by suspension in PBS containing 10g / L agarose in serum-free medium, adjusting the concentration to make every 10μl containing 1×106 C6 glioma cells and placed them into 37oC water bath to be vaccinated.For the establishment of a stable and reliable model of rat brain C6 glioma, using stereotactic instrument planting C6 glioma cells, the establishment of C6 glioma rat model 35, were divided into four groups, respectively, on the 7th day (n = 5), the first 14 days (n = 5), No. 21 days (n = 5) line of MRI scans and finally a group of natural death (n = 20) for the survival analysis. With 1.5 T MRI scanner for imaging, for T1-weighted scan (T1WI), T2-weighted scan (T2WI), and enhanced T1WI scans were observed after 1,2,3 weeks after inoculation tumor MRI changes in tumor growth, and with the general pathology. The results were compared.A general clinical observation after Vaccination , and vaccination of rat aorta after execution done paraformaldehyde perfusion fixed brain tissue and tumor biopsy specimens observation and HE staining and GFAP immunohistochemical staining .Results: 1. In this study, the caudate nucleus stereotactic inoculation C6 glioma cells, all rats were inoculated glioma growth (100%), immunohistochemical staining positive for GFAP confirmed that the tumor is glial cell line-source; tumor growth rate in the first started two weeks has accelerated noticeably.2. MRI manifestations of normal rat brain tissue: T1WI phase: was slightly lower signal, unclear delineation of cortex and white matter, ventricles and subarachnoid space and can not tell; T2WI phase: cerebral cortex and white matter can better distinguish the signal above the cortical white matter signal, after the bilateral lateral ventricle angle showed a high signal; bilateral tentorial can better in T1WI and T2WI showed symmetric low signal; T1WI-phase enhanced scan, brain tissue was found to strengthen, but the shadow can be seen to strengthen blood vessels.3. After inoculation in rat C6 glioma cells line the first week T2WI, T1WI enhanced scan can be displayed, you can see the tumor growth and tumor of tumor edema of the images, two weeks after the tumor showed rapid growth of the tumor, tumor size, and general pathology were similar.Conclusion: 1. In this study, stereotactic method to the caudate nucleus of rat C6 glioma cells inoculated in rat glioma model of preparation method, the vaccination rate up to 100%. 2. Clinical 1.5 T MRI T1WI enhanced scan, T2WI can better reflect the changes in rat brain glioma growth, for gliomas in vivo provide the basis for experimental study. Growth of C6 glioma tumor volume o step into the law of the Gamma Knife treatment of glioma provide the basis for experimental study. 3. In the second to the third weekend, the weekend between the lines related to medical treatment is the right time. The second part: Gamma Knife treatment of rat C6 glioma before and after VEGF, AQP1, AQP4 expression changes and significanceObjective:Rat C6 glioma by the gamma knife treatment, testing before and after gamma knife radiosurgery for VEGF, AQP1, AQP4 changes, to explore the mechanism of gamma knife treatment of glioma.Methods: 50 male SD rats were taken, weighing between 350-500g. Stereotactic method of 40 SD rats were animals, modeling, tumors were divided into control group and Gamma Knife treatment group, n = 20, and the remaining 10 normal rats. Gamma Knife treatment 7 d, all rats were killed and immunohistochemistry to detect VEGF, AQP1, AQP4 and analyzed statistically.Results: 1. VEGF basically no expression in normal rats, rats in the control group in tumor expression increased, gamma knife irradiation reduced the expression of VEGF. It indicates that the tumor vessel'growth was inhibited after Gamma Knife treatment. The gamma knife treatment for glioma indeed curative.2. AQP1 in normal brain tissue, almost no expression, AQP4 in normal rat brain tissue to express.3. Gamma-ray irradiation, VEGF, AQP1, AQP4 were reduced to varying degrees.Conclusion: The gamma-ray can reduce VEGF expression in rat C6 glioma, can reduce AQP1, AQP4 expression.
