Objective: To research the mutation of cell-free K-ras gene of the laryngeal carcinoma patients (before operation and one day, five day and ten day after operation). Analyse the correlation between the mutation of cell-free K-ras gene and the clinical factors (eg : TNM clinical classification, clinical stage, pathological grade ) of the patients. To give a theoretic basis on the noninvasive detection of patients before operation, distinguish benign and malignant tumor and prognostic surveillance, then giving a theoretic basis on the early diagnose and treatment.Methods: Collect twenty-nine patients'peripheral blood (before operation and one day, five day and ten day after operation). Take ten volunteers'blood as control. Using Luminex liquichip and mutant-enriched PCR to detect the point mutation of codon 12/13 in K-ras gene in each sample.Results: Difference between the rate of mutation of cell-free DNA K-ras gene in laryngeal carcinoma group (before operation) and the control group, difference between laryngeal carcinoma group (after operation) and the control group, difference between before operation group and after operation group, are not significant (P >0.05). The rate of mutation of cell-free DNA K-ras gene in laryngeal carcinoma group increases with the raise of TNM clinical classification, clinical stage and pathological grade in laryngeal carcinoma group. Difference in each clinical factors is not significant (Fisher'exact test value >0.05).Conclusion: The point mutation detection of cell-free DNA K-ras gene in laryngeal carcinoma patients may not be applied to clinic. Luminex liquichip technology and cell-free DNA detection will have some appliance prospects on clinic.
