Objective:To observe the effect of Feifukang Decoction on the survival status of Lewis lung cancer cell bearing tumor mice and the growth of transplanted tumor.To investigate whether pulmonary reactants can play an anti-tumor mechanism by regulating the expression of Bcl-2,Bax and Caspase-3,and provide experimental evidence for the treatment of lung cancer.Methods:The Lewis lung cancer cell suspension was adjusted to lx107/ml and rapidly inoculated into the right armpit of 50 C57 mice.When the tumor was grown on day 14,50 mice(tumor 150mm3)were observed and selected.50 mice were randomly divided into 5 groups,each group of 10,respectively,for the Feifukang high dose group,Feifukang middle dose group,Feifukang low dose group,cisplatin group,model group.Another normal mice 10 for blank group.Feifukang high dose group:52g/kg 0.4ml gavage for 14 days,Feifukang middle dose group:26g/kg 0.4ml gavage for14 days,Feifukang low dose group: 13g/kg 0.4ml for 14 days,cisplatin group: 2mg/kg 0.2ml intraperitoneal injection once every other 3 times,the model group:0.4ml normal saline for 14 days,blank group: 0.4ml saline for14 days.During the experiment,the mental state of the mice,mouse fur,diet and defecation were observed.The mice were sacrificed at 24 hours after the last administration,and the tumor inhibition rate was calculated.The expression levels of Bcl-2,Bax and Caspase-3 were detected by immunohistochemistry.The level of apoptosis was measured by tunel method.Result:1.Comparison of the quality of life in mice: The experimental group compared with the blank group,the growth、 the diet、 the activities and the survival of the state of mice were reduce,but Feifukang high,medium and low dose group living condition is better,cisplatin group and model group living condition is poor.2.Comparison of the effects on transplanted tumor in mice:The results of each group were as follows(`x±s): high dose of Feifukang group:3.63 ± 0.36 g,middle dose of Feifukang group: 4.34 ± 0.22 g,low dose of Feifukang group: 5.34 ± 0.56 g,cisplatin group: 4.13 ± 0.24 g,model group:6.27 ± 0.45 g.Compared with the model group,the tumor weight in the high,middle and low dose group and cisplatin group decreased,and the difference was statistically significant(P< 0.01).Feifukang high dose group compared with cisplatin group tumor weight decreased and the difference was statistically significant(P< 0.05),Feifukang meddle dose group compared with cisplatin group tumor weight increased but the difference was not statistically significant(P> 0.05),Feifukang low dose group compared with cisplatin group tumor weight increased but the difference was statistically significant the(P< 0.01),the high dose group of Feifukang conpared with Feifukang middle 、 low dose group and meddle dose group of Feifukang compared with low dose group tumor weight decreased and the difference was statistically significant(P< 0.01).3.Effect of tumor inhibition rate: Feifukang high dose group:42.03%,cisplatin group:34.08%,Feifukang middle dose group:30.80%,Feifukang low dose group:14.78%.4.Effects of gene expression on tumor cells:In Bax,Caspase-3 and apoptosis in pulmonary rehabilitation group: high,medium and low dose group and cisplatin group compared with the model group,the expression was increased,which was statistically significant in the Bax group(P< 0.05),with significant statistical significance in Caspase-3 and apoptosis group(P<0.01),the high dose group and cisplatin group expression were increased and was statistically significant(P<0.05),Feifukang middle dose group compared with cisplatin group expression decreased but not statistically significant(P>0.05),Feifukang low dose group compared with cisplatin group was decreased and statistically significant significant(P<0.01),Feifukang high dose group compared with middle and low dose group increased expression and there was statistical significance(P<0.01),Feifukang medium dose group compared with low dose group was increased,but in the apoptosis group was statistically significant(P<0.05),in Bax and Caspase-3 group had significant statistical significance(P < 0.01).In the Bcl-2 group: Feifukang high dose group and cisplatin group compared with the model group,the expression was decreased and there was statistical significance(P<0.01),Feifukang low dose group compared with model group and Feifukang high dose group compared with cisplatin group expression were decreased and the difference was statistically significant(P<0.05),Feifukang middle dose group and cisplatin group expression increase but had no statistical significance(P>0.05),Feifukang low dose group and cisplatin group expression increased and was statistically significant(P<0.01).Feifukang high dose group compared with the middle dose group and low dose group,Feifukang middle dose group compased with lou dose group the expression were decrease and has statistical significance(P< 0.01).Conclusion: Feifukang can improve the survival of mice with Lewis lung cancer and inhibit the tumor growth,the study confirmed that it may inhibit the growth of tumor cells by promoting the expression of Bax,Caspase-3and inhibiting the expression of Bcl-2 related.
