Objective:To detect mRNA expression differences of leptin in 3T3-L1 adipocytes, which was in vitro stimulated under different concentrations of glargine insulin (powder), glargine insulin (liquid), detemir insulin (liquid), human insulin (powder); to detect the leptin mRNA expression differencesin 3T3-L1 adipocyte,which was stimulated with the same concentration of glargine insulin(powder), glargine insulin(liquid), detemir insulin(liquid), and human insulin (powder); To explore the effect of different concentrations of insulin,glargine insulin and detemir insulin on the expression of leptin mRNA in 3T3-L1 adipocytes.Methods:1. To culture 3T3-L1 preadipocytes in vitro, and differentiate into mature fat cells.2. 3T3-L1 adipocytes were incubated with different concentrations of insulin and insulin analogue for 24 hours. According to the different types of induction medium containing insulin were randomly divided into 4 major groups: human insulin (powder) , glargine insulin (powder), glargine insulin (liquid), detemir insulin (liquid); According to each group contained different concentrations of insulin (1,10,100 nmol/L) was divided into 3 groups. A separate control group, the insulin concentration 0nmol/L. Sample size in each group of 3, repeat 3 times.3. To detect the levels of leptin mRNA expression in 3T3-L1 adipocytes by using real time fluorescence quantitative PCR.Results:1. Different concentration (1nmol/L, 10nmol/L, 100nmol/L), different kinds of insulin (human insulin (powder) , glargine insulin (powder), glargine insulin (liquid) and detemir insulin (liquid)) acted on mature 3T3 - L1 adipocytes for 24h, along with the increase of insulin concentration,adiponectin mRNA expression increased gradually,the difference was significant (P<0.05);2. The expression of leptin mRNA in any two groups have statistical significance with the same concentrations of insulin(P <0.05),Except within the two groups ,glargine insulin (powder) and glargine insulin (liquid) group.Conclusion:1. Human insulin (powder), glargine insulin (powder), glargine insulin (liquid) and detemir insulin (liquid) in vitro promote leptin mRNA expressionin in 3T3-L1 adipocytes. The leptin mRNA expressionin was increasing with more concentration. 2. Human insulin (powder) play the strongest role in leptin expression in 3T3-L1 cells,followed by glargine insulin (powder) and glargine insulin (liquid). Detemir insulin (liquid) was the weakest.
