As a malignant tumor, lung cancer has the biological characteristics of invasion and metastasis. In these processes, tumor cells interactive with basement membrane firstly,which composed of extracellular matrix. Therefore, extracellular matrix is critical to bear the invasion and metastasis of tumor. Matrix metalloprotei-nase family has the ability to degrade extracellular matrix, earning more and more attention. As one of the matrix metalloproteinases, MMP-10 is being significant because of its unique role. It can not only degrade a variety of extracellular matrix, at the same time degrade pro-matrix metalloproteinases and activate them. MVD within the tumor angiogenesis as a judge of indicators, has been widely considered to be an important factor in predicting prognosis of tumor. CD105 is expressed mainly in proliferating endothelial cells, and signs angiogenesis on specific, so become an important marker of MVD. Meanwhile, CD 105 can also be regulating the activity of the TGFβto promote endothelial cell proliferation, differentiation and migration, and participates in angiogenesis.In present, there is little research about the relation of MMP-10 and non-small cell lung cancer, which reveal the role of MMP-10 of development, invasion, metastasis and angiogenesis. And there is no reported about the interaction of MMP-10 and MVD (marked by CD105) in non-small cell lung cancer. This research, we detected MMP-10 and MVD (marked by CD105) expression in non-small cell lung cancer by immunohistochemical methods, And analyzed relationships among MMP-10, MVD and lung carcinoma, discussed the interaction between MMP-10 and MVD(marked by CD105) in lung carcinoma, aiming at supplying diagnosis and prognosis indicator and new therapy target site for lung carcinoma.MethodsSamples of lung cancer and adjacent normal lung tissue were collected from 60 patients with NSCLC, diagnosed by pathology of surgical removed tissues from 2006 to 2008 in the First Affiliated Hospital of Zhengzhou University.20 normal lung tissues were collected as control.All the patients were not received treatments including chemotherapy, radiotherapy and others. Among them there were 42 males and 18 female, aged from 28 to 75 years old and the average age is 55.6. There were 22 poorly-differentiated cases,18 moderately-differentiated cases,20 well-differentiated cases;28 squamous cancer cases,32 adenocarcinoma cases by pathological classification. Lymph node metastasis was observed in 17cases, and no lymph node metastasis in 43 cases. Twenty-eight cases are inⅠandⅡclinical stage, 32 cases are in III and IV clinical stage; Samples were fixed in 10% neutral formalin and embedded in paraffin.The expressions of MMP-10 and CD105 in lung cancerous tissue were detected by SP immunohistochemical method. The data were analyzed by software SPSS10.0. Chi-square method is used to compare the difference of MMP-10 expression between groups. T-test is used to compare the expression of MVD(marked by CD105) between groups, and assess the relation of MMP-10 and MVD.Results(1). Expression of MMP-10 in NSCLC:The positive rate of MMP-10 was 73.3% in samples of NSCLC tissues and 30% in normal lung tissues (P<0.05). The positive rate of MMP-10 in samples of well-differentiated and moderately-differentiated cases was 63.2%, lower than the expression in poorly-differentiated lung cancer(90.9%)(P<0.05).The positive rate of MMP-10 in the group with lymph node metastasis was 83.8%, higher than the expression in the group without lymph node metastasis(56.5%)(P<0.05). The positive rate of MMP-10 in samples of I and II clinical stage cases was53.6%, lower than the expression in poorⅢandⅣclinical stage lung cancer(84.4%)(P<0.05). There were no statistical significance between squamous cancer and adenocarcinoma, male and female, smoker and non-smoker, younger and older. (2). Expression of MVD (marked by CD 105) in NSCLC:MVD value of non-small cell lung cancer was 43.75±3.97, normal lung tissue was 33.25±3.31, the difference between their was significant (P<0.05). MVD value of well-differentiated and moderately-differentiated cases was 39.6±5.1, lower than the expression in poorly-differentiated lung cancer(44.7±7.3)(P<0.05). MVD value I and II clinical stage cases was37.6±5.4 lower than the expression in poorⅢand IV clinical stage lung cancer(44.9±7.5)(P<0.05). MVD value the group with lymph node metastasis was 43.8±6.1, higher than the expression in the group without lymph node metastasis(37.8±5.5)(P< 0.05). There were no statistical significance between male and female, smoker and non-smoker, younger and older, squamous cancer and adenocarcinoma. (3). The relationship between MMP-10 and MVD(marked by CD105) in NSCLC:MVD value of MMP-10-positive group (39.23±2.6) was higher than MMP-10 negative group (32.38±3.8). There was significant difference between the two groups (F=6.988 P=0.013)Conclusion(1). The levels of the expressions of MMP-10 were closely related to lymph node metastasis, differentiation grade, clinical stages (P<0.05). MMP-10 may be involved in the development, invasion, metastasis and angiogenesis and other processes of non-small cell lung cancer. (2). MVD (marked by CD 105) value in non-small cell lung cancer correlated with the lymph node metastasis, tumor cell differentiation and clinical stage. This indicated that CD105 may be promote the progress of angiogenesis, invasion and metastasizing of non-small cell lung cancer (3). The expression of MMP-10 and MVD (marked by CD105) in non-small cell lung cancer showed a positive correlation with each other. This indicated that they may interactive in the progress of the occurrence, development, invasion, metastasis and angio-genesis of lung cancer.
