Objective: Recently, we have analyzed biochemistry of gingival crevicular fluid.We have found that the cytokine has an effect of immunoregulation and immediate induction in the destruction pathology of periodontitis.The cytokine may be an index of diagnostics or periodontal inflammation.In the experiment,the content of Interleukin-23 and keratinocyte growth factor-1 in healthy periodontal sites and in periodontitis sites before and after periodontal treatment were measured by enzyme-linked immunosorbent assay(ELISA). The relationship was discussed among Interleukin-23 and keratinocyte growth factor-1 in GCF and clinical parameters. The purpose of the study is to research the relationship among keratinocyte growth factor-1 and Interleukin-23 are regarded as beneficial indexes to reflect the periodontal status or not;and to explore the effect of Interleukin-23 and keratinocyte growth factor-1 in periodontitis pathology and the illness progress. Methods: 30 subjects who had periodontitis in the study were selected as the case group,30 healthy volunteers who had no periodontal disease were selected as the normal group. The same molar tooth was selected as the study object from all the subjects. The case group was devided into pretherapy and post-treatment(the fourth week,sixth week and the eighth week) through periodontal initial therapy,to observe and record the clinical parameters:SBI,PD,AL of every selected tooth.GCF was collected by paperstrip,ELISA was used to detect the levels of keratinocyte growth factor-1 and IL-23 in GCF,and an correlation check was done between the clinical parameters and the concentration of KGF-1 and IL-23.Results:⑴The concentration of KGF-1 and IL-23 in GCF in the inflammatory group was significant higher than the normal group(P<0.05), at the fourth week,sixth week and the eighth week after the treatment,the concentration of KGF-1 and IL-23 in GCF was lower than inflammatory group before treatment(p<0.05). and significant deference of KGF-1 and IL-23 concentration and statistical significance were found in normal group and in different stages after treatment(P<0.05).⑵The clinical parameters showed significant deference between the inflammatory group and the normal group or pretherapy and post-treatment.⑶Strong positive relationships were found between the level of KGF-1 in GCF and the clinical parameters(SBI,PD,AL); The correlation coefficient were 0.545,(p=0.039); 0.617,(p=0.012);0.537, (P=0.031);strong positive relationships were found between the level of IL-23 in GCF and the clinical parameters; The correlation coefficient were 0.467,(0.031); 0.629,(0.019);0.541,(0.027).Conclusion: There existed KGF-1and IL-23 in GCF of periodontitis patients. The concentrations of IL-23 and KGF-1 in periodontitis sites were significantly higher than in healthy sites. And the concentrations of IL-23 and KGF-1 at every stage of post-treatment were lower than in periodontitis sites in the inflammatory group. The increase of IL-23 and KGF-1 levels can reflect the degree of the inflammatory periodontal disease, so the two factors played an important role in the cause and development of periodontitis. In summary,we can regarded KGF-1 and IL-23 as sensitive diagnosis indexes of periodontitis and beneficial indexes that reflect the status of periodontitis.
