ObjectiveTo study on whether the Cerulenin can induce the prostate cells PC-3 to be apoptosis.Methods1. Observed with MTT on the PC3 inhibitory effect of cerulenin in vitro.2. Study the PC-3 cell cycle change and apoptosis by using the FCM.3. Study the FAS protein change by using the Western blot.Results1. By the MTT method, the apoptosis rate for PC-3 cells were (34.78±2.47)%, (46.76±3.18)%, (58.13±3.55)%, (65.31±2.81)% by the treatment time on 24h. Compared with the control group was statistically significant (P<0.05)2. In the FCM system the apoptosis rate for the PC-3 cells were 28.29±1.88%,44.73±2.69%,61.25±1.43% in three time groups(24h,48h,72h) under the Cerulenin concentration in 5.0mg/L, and increase in the proportion of cells in G0/G1, S phase and G2/M phase cells decreased.3. Western blot experiments, the longer drug use PC-3 cells less protein, drug action time and FAS protein expression in a negative correlation.Conclusions 1. Cerulenin can inhibit proliferation of PC-3 cells, this inhibition showed a positive correlation with the processing time and the dose of Cerulenin.2. Cerulenin can make an influence in the cell cycle of PC-3 cells, blocking it in the G0/G1 phase and induce apoptosis.
