The Study Of Antiproliferation Effect Of Cerulenin Combined With Epirubicin On Human Osteosarcoma U2-OS Cells

Objection:To investigate antiproliferation effect of Cerulenin combined with Epirubicin on human osteosarcoma U2-OS cells.Methods:1. Epirubicin and Cerulenin, alone or in combinataion,were given vitro to U2-OS cells. The cells proliferation was measured by MTT analysis. The interaction between cerulenin and Epirubicin was assessed using the Jin's Q.2. The Flow cytomety was used to determine the apoptosis induced by Epirubicin or Cerulenin on human osteosarcoma U2-OS cells, then calculation the apoptosis rate.Results:1. Weather Cerulenin or Epirubicin was able to inhibit the proliferation of human osteosarcoma U2-OS cells in vitro based on dose dependence. In combination group, the inhibition rate increased obviously. Q value were 1.24 (when 1μg/ml Epirubicin combined 1μg/ml Cerulenin) and 1.96(when 10μg/ml Epirubicin combined 20μg/ml Cerulenin).2. The Flow cytomety showed that weather Cerulenin or Epirubicin was able to induce apoptosis on human osteosarcoma U2-OS cells.After administrating lμg/ml,5μg/ml,10μg/ml Epirubicin for 24 h, the rate of apoptosis is 17.27%,26.07%,31.54% respectively. After administrating 1μg/ml,5μg/ml,20μg/ml Cerulenin for 24 h, the rate of apoptosis is 7.32%,11.17%,38.78% respectively. After administrating 1μg/ml Epirubicin combined 1μg/ml Cerulenin for 24 h, rate of apoptosis is 26.94%(Q value was 1.17) and 10μg/ml Epirubicin combined 20μg/ml Cerulenin is 66.19%(Q value was 1.17).Conclusion:Weather Cerulenin or Epirubicin was able to induce apoptosis and inhibit the proliferation of human osteosarcoma U2-OS cells. Cerulenin could enhance the inhibition of Epirubicin on human osteosarcoma U2-OS cells.
