Objective: Chronic atrophic gastritis with intestinal metaplasia or hyperplasia namely precancerous lesion of grastic cancer, By establishing the model of PLGC experimental animals, To observe the curative effect of Ying Lian Jin Hua Zhuo Jie Du Decoction on PLGC rats and the influence of on the expression P53 protein, vascular endothelial growth factor (VEGF) and serum PGⅠ, To approach the possible action mechanism about the therapy of this prescription to PLGC, at the same time, to provide experimental evidence for clinical practice.Methods: 80 healthy male Wistar rats of clean grade were randomly divided into 5 groups. Each group had 17 rats, except the normal group has 12 rats: the normal group (NG), the model group (MG), the Chinese medicine high-dosage groups (HDG), the Chinese medicine small dose group (LDG), the Wei Fu Chun group. Except the normal group, the rest groups made the comprehensive methods. The first seventh days after successful modeling, the normal group was given the normal feeding, the model group fed with normal saline. And the other group rats were treated with physic liquor. We observed the changes of mental state, activity, color pattern, appetite, body weight, general changes in stool, etc. Rats in each group were sacrificed after 5 month of gavage, Each rat was taken 5ml of blood to detect the PGⅠ. By immunohistochemical detection, we observed the expression of P53 protein, VEGF on stomach tissue of rats, Observing simultaneously the general view and pathology in gastric mucosal tissue, Effective evaluation based on the observed results, discussing the Ying Lian Jin Hua Zhuo Jie Du Decoction on P53 protein, PGⅠand VEGF effects. Results:1 The general stateCompared with the normal group, the rats of built model 3 weeks began to appear the mental state decreased, hair loose, glossiness poor, the activity decreased, loss of appetite, weight loss. Four weeks later, The HDG, LDG and Wei Fu Chun group of rats generally had different degrees of improvement, including LDG and Wei Fu Chun group had less improved than the HDG in the general conditions of rats, LDG had less improved than HDG, Wei Fu Chun group was not as more improved as LD group in the general conditions, while MG of rats recovered the worst in the general state.2 The general and histopathological observation of gastric mucosal tissue NG rats, a pale red of gastric mucosa, all of the stomach wall thicker, elasticity better, mucosal plica surface smooth, toward the rules modifying the more mucous. HDG, LDG and Wei Fu Chun group were significantly decreased compared with MG (P<0.05). LDG and Wei Fu Chun group were significantly decreased compared with HDG (P<0.05). LDG and Wei Fu Chun group compared have statistically significant difference (p<0.05). With histologic situation of rats, HDG and NG were significant differences which compared with MG (p<0.05), LDG and Wei Fu Chun group, which compared with MG, have not remarkable difference, and the differences have no statistical significance (p>0.05). The expression level of VEGF in HDG, LDG and Wei Fu Chun group were (0.0862±0.0297), (0.1414±0.0271), (0.1871±0.0339). The treated groups were significantly decreased compared with MG (p<0.05). The treated groups and NG have remarkable difference, and the differences have statistical meaning (p<0.05). HDG and LDG have remarkable difference, and the differences have statistical meaning (p<0.05). LDG and Wei Fu Chun Group have remarkable difference, and the differences have statistical meaning (p<0.05).4 Effect of Ying Lian Jin Hua Zhuo Jie Du Decoction on PLGC Model Rats'the content of serum PGⅠCompared with NG, MG, LDG, HDG, Wei Fu Chun group have remarkable difference, and the differences have statistical significance (p<0.05). Namely each group the content of serum PGⅠwas relative lower than NG (99.48±4.27). MG (52.00±3.33) compared with HDG, LDG and Wei Fu Chun group, there was a statistically significant (p<0.05). HDG (85.53±1.83) compared with LDG (71.30±2.43) and Wei Fu Chun group (62.27±2.20), the differences have statistically significant (p<0.05). There were different degrees of improvement, for example: mental status, mobility, appetite, weight, fur color, etc. At the same time, it could also improve gastric mucosal inflammation of the situation, intestinal metaplasia and hyperplasia, so fundamentally reversed intestinal metaplasia and hyperplasia. The study results showed that these related to a dose-dependent efficacy of traditional Chinese medicine.2 Ying Lian Jin Hua Zhuo Jie Du Decoction from the level of molecular biology research its mechanism, It could cut p53 protein expression level, inhibiting cell proliferation and transformation, making the cells stop growing and inducing apoptosis and exert its function of PLGC. It might also reduce the level of VEGF expression, Thereby reducing endothelial cell proliferation. Meanwhile, research showed that it could raise the level of serum PGⅠexpression, then reversing CAG especially associated with intestinal metaplasia and hyperplasia.
