Objective:The present series of experiments were performed to observe the changes of serum tissue-type plasminogen activator and plasminogen activator inhibitor-1 after massive cerebral infaction who undertake therapeutic hypothermia. And to explore the effect of therapeutic hypothermia on serum tPA/PAI-1 activity of the massive cerebral infarction patientsMethods:46 Patients who sufferd from acute massive cerebral infarction and whose insults must be on within 48 hours were randomly divided into conventional treatment group and hypothermia group.The hypothermia group subjects accepted conventional treatment and hypothermia therapy. To detected the serum tPA and PAI-1 activity when the patients arrived at hospital and 48 hours after treatment respectively. At the meantime, we also detected the the serum tPA and PAI-1 activity as control.Results:1,The activity of serum tPA and PAI-1 are significantly increased after acute massive cerebral infarction from 56.1±32.51pg/ml,121.1±35.44pg/ml to 132.7±40.25pg/ml, 134.5±45.74pg/ml; 207.0±42.04pg/ml,206.4±42.30pg/ml (P <0.05).2,After 48 hours'convertional treatment, the activity of serum tPA and PAI-1 were significantly decreased to 110.1±32.65pg/ml,174.2±38. OOpg/ml (P<0.05). However, the activity of serum tPA and PAI-1 of hypothermia group were significantly decreasd to 81.0±34.64pg/ml,145.4±45.29pg/ml (P<0.05).After 48 hours, the NIHSS score of convertional treatment group was 30.1±6.17, while the hypothermia group was 24.9±5.59 (P<0.05)Conclusions:1,The activity of serum tPA and PAI-1 are significantly increased after acute massive cerebral infarction.2,Therapeutic Hypothermia could lower the activity of serum tPA and PAI-1.3,Therapeutic hypothermia could play neuroprotective effrcts by decreaseing the activity of serum tPA,PAI-1 which also provides theoretical basis for its clinic practice.
