Expression, Purification and Characterization of sTRAIL-TMTP1 FusionsObjective:. Using prokaryotic expression system purified fusion protein sTRAIL-TMTP1, study the effects on apoptosis of tumor cell lines in vitro, observe its biological effect of prostate cancer subcutaneous tumor model and gastric cancer orthotopic model in vivo. Measures: Application of PET prokaryotic expression system and purification of fusion protein sTRAIL-TMTP1; Applications Western Blot and Coomassie blue staining fusion protein; Using flow cytometry for effect of apoptosis by fusion protein on tumor cell; establishe prostate cancer cell line PC -3M-1E8 subcutaneous tumor model, peritumoral injection of fusion protein treatment; the establishment of gastric cancer MKN-45sci in situ model, intraperitoneal injection of fusion protein treatment; Using quantum dots, and immunohistochemistry to check drug metabolism and tumor tissue and other organ cells apoptosis.Results: By purified protein is as enough as be detected by antibodies;Protein can obviously lead to apoptosis of high metastatic tumor cells; Effect on tumor of fusion protein is obviously in vitro, tumor volume and number of tumor metastasis. Conclusion: The fusion protein sTRAIL-TMTP1 has a good anti-tumor effect both in vivo and in vitro. Effect of GDF-5 on tendon cell and sheath cell of rat AbstractObjective: To explore the effect of growth differentiation factor-5(GDF-5) on tendon cell and sheath cell of rat, and lay the foundation for our further research on enhancing healing of hurt sheath by GDF-5 and improve adhesions by endogenous healing in vivo. Method: The proliferation rate of tendon cell and sheath cell was investigated with MTT assay,while the apoptosis rate of tendon cell and sheath cell was evaluated by using Flow Cytometer. Resμlts: GDF-5 can precipitate the proliferation rate of tendon cell obviously and stimμlate the proliferation rate of sheath cell a little, the impact relates to the concentration of GDF-5, but it does not impact their apoptosis rate. Conclusion: Some concentration of GDF-5 can precipitate the proliferation rate of tendon cell more than sheath cell, while it has little impact of their apoptosis.
