Experimental Study On The Shuangshen Tianqi San On Model Rats With Atherosclerosis

Objective:To observe the anti- atherosclerosis effects of Shuang shen tian qi san,SSTQS for short, a chinese herb compond,on treating atherosclerosis(As)of modle rats,trying to investigate the mecchanism of it's therapeutic effects.Methods: Rat model with atherosclerosis was established by feeding fatty forage and vitamin D3 administration(vitamin D3600,000IU/kg by celiac injection at one time) for 12 weeks.52 healthy male Sprague-Dawlay rats were randomly divided into 4 groups: 1.normal group ( feed with normal diet);2.model group(feed with fatty-diet);3.SSTQS group.(feed with fatty-diet+SSTQS:1g/kg.wet.d).4. Simvastatin group(feed with fatty-diet+ Simvastatin:4mg/ kg.wet.d).The SSTQS or Simvastatin was fed by gavage to the SSTQS and Simvastat in group, mice for 12 weeks.the normal group and model group were given the same volume of saline as SSTQS group.By the end of 12th weeks, The serum lipid was contented ,and killed the rats.After the rats were killed,blood was collected to mearsure the levels of serum total eholesterol(TC),low density liPoProtein cholesterol (LDL-C),high density liPoProtein cholesterol (HDL-C),triglyeeride(TG), malonaldehyd(MDA),suPer oxide dismutase(SOD). the changes in the aortas'ultrastructure were to be observed through the electron microscope.Results:(1)With SSTQS treatment, Pathomorphological changes in the aorta:the pathological changes lessened in the SSTQS group,compared to the model group. The aorta tissue obviously improved. (2)the activity of SOD increased lighly;the level of serum MDA was obviously decreased,compared to the model group(P<0.01).(3)contents of serum TG(P<0.05),TC(P<0.01),LDL-C(P<0.01)of atherosclerosis rats were significantly decreased ,compare to the model group.Conclusion: SSTQS have therapeutic effects on atherosclerosis of rats, including:(1)Morphological changes may suggest the SSTQS can protect the aorta endothelial cell,improve the pathological change in aorta.(2)SSTQS has the effect on regulating blood-lipid metalolic disorder .(3) decrease the serum levels of MDA,and increas the Serum level of SOD on AS rats, the therapeutic effects of SSTQS maybe through Preventing lipid Peroxidation.(4)The experimental model of atherosclerosis in rats may be conveniently established by loading of high fat diet and vitamin D3 600,000IU/kg by celiac injection at one time),which can be used for study of atherosclerosis.
