Panax notoginseng,also known as "Panax pseudo-ginseng" and "Radix notoginseng","Dianqi","Shanqi",is a bright pearl of traditional Chinese medicine treasure house.It was originally recorded in the "compendium of Materia Medica",with the effect of bleeding,blood stasis,swelling and analgesic therapy.is known as good medicine on hemorrhagic disease and effective medicine on department of traumatology.Panax notoginseng is one of the rare Chinese herbal medicines,many books are on Panax notoginseng for the full affirmation,have the"invaluable" and "Southern God grass","the king of ginseng" reputation and so on.Raw and steamed panax notoginseng have goog function and taste but exist significant differences,Chinese medicine and folk have notoginseng "shengdashubu" said that it has been in Panax notoginseng and processed products of the two kinds of form is used in clinic,pharmacological action and clinical indications of the two forms are different and the adulteration are more and more serious,so strict quality control is very important.there are currently many researches about pharmacological actions and chemical composition analysis of panax notoginseng around,but few studies on processed products of panax notoginseng,processing mechanism of panax notoginseng and hematopoiesis mechanism are still unable to get a reasonable explanation,the relationship between material basis of panax notoginseng "shengdashubu" and efficacy similarities and differences of raw and steamed panax notoginseng needs to be further researched.Therefore,in order to promote the further development of notoginseng industry,expand its application scope,there are important significance that taking in-depth study on material basis of panax notoginseng " shengdashubu" and clarifying the processing mechanism and mechanism of action,it helps to develop more stringent ang regulatory quality standards and the use of norms of raw and steamed panax notoginseng,and is conducive to realize the modernization and internationalization of Chinese medicine.In this paper,the contents of various chemical constituents in raw panax notogindeng and different processed products were compared,including the saponins,dencichine,total flavonoids,amino acids and soluble sugar.In order to obtain the effective part saponins group of panax notoginseng,industrial procession,it used the heat extraction and macroporous resin purification to get steamed panax notoginseng total saponin,and then discusses pros and cons about separation methods of effective fractions of steamed panax notoginseng saponins by the silica gel column chromatography and semi manufactured liquid chromatography,finally compared hematopoietic activity of raw and steamed notoginseng,effective fractions of steamed panax notoginseng saponins on blood deficiency model mice that induced by compound use of cyclophosphamide and acetyl phenylhydrazine.The research content of this paper included the following:1.Processing technology research of steamed panax notoginseng.The steaming method was used for processing panax notoginseng,the panax notoginseng taproot,fibrous root and rhizome respectively under 105?,110?,120 ? for steaming 2h,4h,6h,8h,10h.Before processing,operation in keeping fresh,dry,and slice of different ways.2.Study on chemical composition variation of raw and steamed panax notoginseng.The effects of different steam temperature and time on the contents of total saponins,monomer saponin,soluble sugar,soluble sugar,dencichine,total flavonoids and amino acids in raw panax notoginseng and different processed products were studied.The results showed that saponins,soluble sugar,soluble sugar,total flavonoids and amino acids and other components of raw panax notoginseng were changed after processed.In addition,it also discussed the effect of different drying methods on saponins content in the steamed panax notoginseng root slices.The results showed that different drying methods have effect on five kinds of saponins of steamed panax notoginseng root slices.In this experiment,raw panax notoginseng and different processed products were identified by Fourier infrared spectroscopy.The resulting from infrared spectrogram showed that the main components of raw panax notoginseng were affected by the processing technology,and the content of glucoside,polysaccharide and other substance in panax notoginseng was reduced after processing,and that was basically consistent with determination and analysis of the content of each component.These results provided certain basis for the study of the correlation between the mechanism of panax notoginseng"shengdashubu" and chemical ingredients change.3.Study on the extraction and purification of total saponins from steamed panax notoginseng.USing the steaming method(about 100?)to pilot-scale processing for panax notoginseng taproot(40kg),that obtained crude steamed panax notoginseng powder after drying and crushing.Then it used the heat extraction to extract total saponins from steamed panax notoginseng,about 8 h,got total saponin extract of steamed panax notoginseng.Then the extract was purified by D101 macroporous resin to obtain steamed panax notoginseng total saponin extract powder(about 4.05kg).After calculation,the yield is 11.52%.4.Study the separation method of the effective parts of steamed panax notoginseng saponins.To find the best method to isolate the effective fractions of steamed panax notoginseng saponins,the results showed that the separation effect of semi preparation liquid chromatography to obtain the effective fractions of steamed panax notoginseng saponins was well.The silica gel column chromatography has the advantages of no limit on the amount of sample,but the operation is complex,the separation effect is not good.The separation effect of semi preparation liquid chromatography method is good,and large polar saponins effective parts and small polar saponins effective fractions of steamed panax notoginseng were separated on the whole.The deficiency of this method is that the sample capacity is limited,and the organic reagent consumption is relatively large.According to the HPLC chromatograms,we chose the semi preparation liquid chromatography method to separate the effective fractions of steamed panax notoginseng saponins.5.The study that blood effect of raw and steamed panax notoginseng and effective fractions of steamed panax notoginseng saponins.This study adopted compound use of cyclophosphamide and acetyl phenylhydrazine in modeling,compared the hematopoietic action of raw and steamed panax notoginseng and effective fractions of steamed panax notoginseng saponins on blood deficiency model mice.Mainly study the influence of peripheral blood of blood deficiency mice,including determination of red blood cell count(RBC),hemoglobin(HGB),red blood cell hematocrit(HCT),mean hemoglobin concentration(MCHC).The results showed that the effect of enriching the blood of steamed panax notoginseng is better-than raw panax notoginseng,due to the existing in the experiment deviation,dosage and drug days are too short and other factors,the effect is not obvious.While steamed panax notoginseng total saponin extract has better hematopoietic activity than raw and steamed panax notoginseng,it explains that saponins are related with blood effect.In.addition,it was found that steamed panax notoginseng total saponin extract has better hematopoietic effect than large polar saponins effective parts and small polar saponins effective fractions of steamed panax notoginseng,which may be illustrated that steamed panax notoginseng with hematopoietic activity was mainly saponins(including part of the original saponins and the new generation of rare ginsenoside)overall in action,and perhaps also related with change of other material composition,that need to further study and validation.
