Research Of Panax Notoginseng And Its Effective Components To DIC Induced By Endotoxin

The dried root of Panax notoginseng which belongs to the plant of Radix Ginseng category (Araliaceae perennial panax species) is a traditional Chinese herb, it can promoting blood circulation to remove blood stasis,reducing swelling and and rlieving pain,it is used principally to treat hemoptysis,traumatic injury and so on; The dried root of Panax notoginseng which was processed can benefite qi and promoting blood circulation, it is used to treat the lost of blood and anemia. The studies about the modern pharmacologic action are furth continually with the technologic development and it indicated that effective components of Panax notoginseng can prevent coronary artery disease, anti-inflammation, anti-aging, anti-oxidation, anti-radical injury, anti-endotoxin and so on. In this experiment, the extraction, identification and medicinal preparation of Panax notoginseng and its effective components and the action of Panax notoginseng and its effective components on Disseminated Intravascular Coagulation (DIC) induced by endotoxin were studied..The first,the best extraction technology were obstained that the dencichine was extractd by optimized means of n-Butanol and the totol Panax Notoginseng was extracted by optimized means of acetone-ether. The quality of injection which was prepared by dencichine,total saponins of panax notoginseng and the was studied. The result showed that the optimum extracting process for radix notoginseng saponin,total saponins of Panax Notoginseng respectively were reflux extraction Methanol was taken as percolation solvent(800ml, for 24h) and then extracted 5 times in n-Butanol,2 times (90min per time) in ethanol. The proportion of acetone:ether=1:1.And the quality analysis showed that radix notoginseng saponin injection was colorless and clearing liquid, pH=7.2, content of radix notoginseng saponin was 100mg/ml,the total saponins of panax notoginseng injection was light yellow and clear liquid, pH=7.4, the content of total saponins of panax notoginseng was 100mg/mL,the water decotion of panax notoginseng injection was light yellow and clearing liquid, pH=7.2,the content of panax notoginseng (dried root) was 1.0g/mL.Secondly, The results of experiment of antibacterial and antiendotoxin in vitro indicated that the Panax notoginseng and its active ingredients were inhibitory effect on bacteria,Panax Notoginsen and total saponins of Panax Notoginseng can inhabit the endotoxin,but dencichine can not inhabit the endotoxin in vitro. Panax notoginseng and its active ingredients were used to cure the patho-model of DIC and judge the effect of treatment. The result indicated that Panax Notoginsen and total saponins of Panax Notoginseng can effectively improve the rabbits' physiological indicators of blood induced by endotoxin,reducing the plasma endotoxin levels,increase the activity of anti-thrombin,prevent and cure effectively the damage of blood vessel induced by endotoxin, the total saponins of panax notoginseng injection was the best.Thirdly,A series means have been used to observe the antiexdotoxin mechanism of total saponins of panax notoginseng and water decotion of panax notoginseng, such as cultured and passaged method of vascular endothelial cells are taken to determinate the activitiesof antithrombin, PAI-1 protein and TF. The results pointed out that the endotoxin can decreased the activity of antithrombin while increased activities of PAI-1 protein and TF in vascular endothelial cells. Total saponins of the Panax notoginseng exerted a considerable influence which depended on the duration and dosage.In summary,this study showed that the total saponins of panax notoginseng and the effective component of Panax notoginseng injecction inhibited the growth of E.coli, Staphylococcus aureus and salmonella, eliminated the negative changes in the part of the blood physiological index induced by endotoxin in rabit effectively, sustained the contents of RBC, WBC and PLT in a normal range and prevented the injury of aortic artery and viscera. In addition, it inhibited the endotoxin induced activity decreased the antithrombin and activities,increased PAI-1 protein and TF as well as its mRNA's expression in vascular endothelial cells and sustained the balance between anticoagulant and procoagulant substances, so the function of vascular endothelia was stable.
