1 PURPOSE Steamed Panax notoginseng is a common clinical decoction piece.Its traditional processing method is atmospheric steaming,which takes a long time.In order to meet the needs of large-scale production and realize high-quality and high-efficiency production,this study explores the standardized processing technology for preparing steamed Panax notoginseng by high-pressure steaming,and makes a comparative study on the chemical components and main drug effects before and after processing.To determine the processing parameters of steamed Panax notoginseng,to clarify the differences of components of raw and steamed Panax notoginseng and the differences of efficacy in enriching blood and promoting blood circulation,so as to provide reference for the production of steamed Panax notoginseng and accurate clinical medication.2 METHODS2.1 Study on Processing Technology of steamed Panax notoginseng Based on the standard of steamed Panax notoginseng produced by traditional normal pressure steaming method,high pressure steaming was used to investigate different steaming temperatures,and the best processing technology of steamed Panax notoginseng was comprehensively judged by taking appearance characteristics and saponin content as indexes.2.2 Comparative Study on Chemical Constituents of Raw and Ripe Panax notoginseng The content of saponins and monosaccharides in raw and steamed Panax notoginseng was determined by high performance liquid chromatography.The content of polysaccharides in raw and steamed Panax notoginseng was determined by ultraviolet-visible spectrophotometry.2.3 Comparative Study on Blood Enriching and Blood Activating Effects of Raw and Mature Panax notoginseng KM mice were divided into 9 groups: normal group,model group,positive drug group,raw Panax notoginseng suspension group,raw Panax notoginseng saponin group,raw Panax notoginseng polysaccharide group,steamed Panax notoginseng suspension group,steamed Panax notoginseng saponin group and steamed Panax notoginseng polysaccharide group.The hemolytic blood deficiency mouse model was established by subcutaneous injection of acetophenone hydrazine.Blood routine examination and pathological sections of liver and spleen were used to compare and analyze the difference of blood enriching efficacy of raw and steamed Panax notoginseng.SD rats were divided into the same groups.The acute blood stasis rat model was established by subcutaneous injection of epinephrine hydrochloride injection combined with ice-water bath.Taking the whole blood shear rate and plasma viscosity as indexes,the difference of blood circulation promoting efficacy between raw and steamed Panax notoginseng was compared and analyzed.3 RESULTS3.1 Processing Technology Reform of Steamed Panax notoginseng Panax notoginseng reached the traditional standard when steamed at normal pressure for 15 hours.The sample was dark brown,completely penetrating,and had no splinters after slicing.Steam at 110? for 3 hours under high pressure,the appearance of the sample reached the traditional standard,and the conversion of saponin components was consistent with that of steam at normal pressure for 15 hours.The high pressure steam processing technology of steamed Panax notoginseng was determined as follows: steam at 110? for 3 hours.3.2 Comparative Study on Chemical Constituents of Raw and Steamed Panax notoginseng Compared with raw Panax notoginseng,steamed Panax notoginseng has added ginsenoside Rk3,Rh4,20(S)-Rg3,20(R)-Rg3,Rg5,etc.However,the contents of notoginsenoside R1,ginsenoside Rg1,Re and Rb1 are significantly reduced to 32.25% ~ 54.5% of the original level.The content of ginsenoside Rd was reduced to 70.8%.Ginsenosides Rc,20(S)-Rh1,20(R)-Rh1 increased 135.2% ~ 826.0% of raw Panax notoginseng.The total amount of ginsenoside Rk3,Rh4,20(S)-Rg3,20(R)-Rg3 and Rg5 newly generated components is 3.325%,accounting for 37.1% of the total amount of steamed Panax notoginseng saponins.Compared with raw Panax notoginseng,the glucose content in steamed Panax notoginseng increased 188.0%,fructose content increased 317.2%,maltose content decreased to the original 40.5%.After processing,monosaccharide(glucose and fructose)content increased and disaccharide(maltose)content decreased.The polysaccharide content of raw Panax notoginseng is 9.264%,that of steamed Panax notoginseng is 12.43%,and that of steamed Panax notoginseng is higher than that of raw Panax notoginseng.It may be beneficial to the dissolution of steamed Panax notoginseng polysaccharide after high pressure steaming.3.3 Comparative Study on Blood Enriching and Blood Activating Effects of Raw and Steamed Panax notoginseng The blood enriching efficacy showed that compared with the model group,red blood cells and hemoglobin in each administration group were significantly increased(P<0.01),hemosiderin deposition and inflammatory cell infiltration in liver and spleen were relieved,and the steamed Panax notoginseng group was superior to the corresponding groups of raw Panax notoginseng.The action intensity of the steamed Panax notoginseng groups is suspension group > polysaccharide group > saponin group.The effect of promoting blood circulation showed that compared with the model group,the whole blood shear rate and plasma viscosity of each administration group were significantly reduced(P<0.01),and the raw Panax notoginseng group was better than the steamed Panax notoginseng group.The effect of raw Panax notoginseng group was suspension group > saponin group > polysaccharide group.4 CONCLUSION In this experiment,the best technological parameters of high-pressure steaming were determined through research,which greatly improved the value-added efficiency.The effect of Panax notoginseng before and after processing changed obviously.In terms of blood enriching effect,steamed Panax notoginseng was superior to raw Panax notoginseng,and the suspension of steamed Panax notoginseng powder had the strongest effect,followed by polysaccharide fraction.In promoting blood circulation,raw notoginseng is better than steamed notoginseng,and the suspension of raw notoginseng powder has the strongest effect,followed by saponin.The results showed that the effects of raw and steamed Panax notoginseng were obviously different,and the traditional powder form was the best for oral administration,which provided a reference for clinical accurate medication.
