OBJECTIVE:To investigate the expression of SPARC in human bladder cancer and to clarify the relationship between the expression levels and the clinicopathological parameters of bladder cancer.METHODS:The expression of SPARC was evaluated by immunohistochemistry in paraffin-embedded tissues from 60 cases of bladder cancer tissue and 20 cases of normal bladder tissue. The correlation between the expression of SPARC and its clinicopathologic features was analyzed accordingly. the results were analyzed with SPSS 13.0 statistical software. a=0.05 was regarded as the statistical criterion.RESUITS:the positive rate of SPARC in cancer group was 80.0%(48/60), significantly higher than that in control group[20%(4/20)], there was significant difference between these two groups (P=0.000). Moreover, The positive rate of SPARC was 93.9%(31/33) in the bladder cancer specimens whose infiltrative depth reached muscular layer, respectively significantly higher than that in the bladder cancer specimens whose infiltrative depth limited mucosa layer or submucosa [63.0%(17/27)],there was significant difference between these two groups (P=0.009). There was more SPARC expression in the patients with metastatic lymph node of bladder cancer (19/20,95.0%) than that without metastatic lymph node (29/40,72.5%), there was significant difference between these two groups (P=0.047).the positive expression rates in gradeâ… ,â…¡andâ…¢were 59.1% (13/22),90.0%(18/20) and 94.4%(17/18),respectively. there were significant differences among the three groups (p=0.007). The SPARC was not related to age,sex (P> 0.05).CONCLUSION:we fistly demonstrated the high expression of SPARC in bladder cancer tissue and the SPARC expression levels positively correlate with the histological differentiation, clinical degree, metastatic lymph node. Our results indicate that SPARC can be of great value in predicting the tumor growth, recurrence, metastasis and poor prognosis of bladder cancer patients. It may be a potential target for bladder cancer.
