Experimental Study Of Effect Of Polyporus Decotion Add Invigorating Kidney Qi Drugs On Rats With Messangial Proliferative Glomerulonephritis

Objective:In this study, Model of mesangial proliferative glomerulonephritis (MsPGN) was made with immunization.To observe the therapeutic effectiveness and mechanism of Polyporus decotion add invigorating kidney qi drugs on MsPGN. Polyporus decotion add invigorating kidney qi drugs compared with Polyporus decotion in treatise, so Polyporus decotion add invigorating kidney qi drugs is more effective prescription to treat MsPGN.Methods:40 healthy wistar rats,were divided into four grous of including normal control group,pathological model group, Polyporus decotion treatment group,Modified Polyporus decotion treatment group. These rats which were removed left kidney except normol group and established by inject BSA and LPS.We gave different medicine.After four weeks,we observe Urine RBC,12h urinary protein,BUN and Cr.Each group of the mesangial cell(MCs) and the extracellular matrix(ECM) were observed with light microscope.The expression of a-SMA and TGF-β1 were observed with the immunohistochemical staining.Result:1.The Urine RBC,12h urinary protein,blood urea nituogen(BUN) and creatinine(Cr) in the model group are higher than the normol group (P<0.01); The expression of a-SMA,TGF-β1 increased in modelgroup,Nephridial tissue pathomorphology showed that:In model group,the mesangial region expanded obviously, mesangial cell(MCs) increased, and the extracellular matrix(ECM) increased.2.Compared with model group,the Urine RBC,12h urinary protein,blood urea nituogen(BUN),creatinine(Cr) and the expression ofα-SMA,TGF-β1 was diminution in treatment group (P<0.01). The above pathology change of two treatments groups obviously reduces, MCs and ECM remarkable decreased.3.There was significant difference between the two treatment group(P<0.05).Conclusions:1.Polyporus decotion add invigorating kidney qi drugs and Polyporus decotion can significantly reduce MsPGN rat's Urine RBC,12h urinary protein,BUN and Cr,improve the change of glomerular pathology to protect renal function.2.Polyporus decotion add invigorating kidney qi drugs and Polyporus decotion can inhibit the expression of a-SMA in nephridial tissue, refrain MsPGN rat's MC proliferation.3. Polyporus decotion add invigorating kidney qi drugs and Polyporus decotion can inhibit the expression of TGF-β1 in nephridial tissue, reduce ECM deposition and delay process of MsPGN.Possibly it is the mechanism of Polyporus decotion add invigorating kidney qi drugs on MsPGN.4. Polyporus decotion add invigorating kidney qi drugs is superior to Polyporus decotion.
