ObjectiveTo investigate expression of nuclear factor(NF)-κB-p65 and matrix metalloproteases-1(MMP-1)and CD15s in lung cancer, and to explore its relationship between the expression and tumor cell metastasis and infiltration and tumor apoptosis,and to explore the possible molecular mechanism of lung cancer cell metastasis and infiltration.Methods1. 60 samples of lung cancer tissues and 15 normal lung tissues after surgical ablation were collected.RT-PCR method was used to detect the expression of NF-κB-p65mRNA in lung cancer tissues and lung tissues around tumor,and the difference in the two groups was analyzed.2. Lung tissues collected were embedded with paraffin, the expression of MMP-1 and CD15s was detected in lung cancer tissues and lung tissues around tumor with Immunohistochemistry. The relationship between the expression of NF-κB-p65mRNA and MMP-1 and CD15s protein was analyzed3. Cell apoptosis index in tumor tissue of lung cancer and lung tissues around tumor was detected by the TUNEL method,and the correlation between the expression of NF-κB-p65mRNA and correspondingapoptosis index was analyzed in lung cancer tissues.Results1. The result of RT-PCR method demonstrated the expression of NF-κB-p65mRNA in lung cancer tissues was significantly higher than that in lung tissues around tumor(0.43±0.11vs0.24±0.06, P<0.05).The expression of NF-κB-p65mRNA in adencarcinoma tissues was higher than that in small cell lung cancer and squamous carcinoma tissues,the expression of three groups had significantly difference(P<0.05). The expression of the NF-κB-p65mRNA was associated with the TNM staging,tumor cell differentiation,the lymph node metastasis and pathological typing,but not associated with gender and age.2. The result of Immunohistochemistry demonstrated that the positive expression of MMP-1 was 34 in 60 cases of lung cancer tissues,its positive rate was 56.7%. The positive expression of CD15s was 36 in 60 cases of lung cancer tissues,its positive rate was 60.0%, but the expression of MMP-1 and CD15s in 15 cases of lung tissues around tumor was negative. The results were statisitically significant(P<0.05). The expression of MMP-1 and CD15s protein was associated with tumor cell differentiation,the lymph node metastasis and pathological typing,but not associated with gender and age. It was statisitically significant.The difference between the two protein was that expression of MMP-1 was associated with the TNM staging,while expression of CD15s was not associated with the TNM staging. By one-factor analysis of variance analysis, the expression of NF-κB-p65mRNA was respectively correlated with the expression of MMP-1 and CD15s in lung cancer tissues.3. The result of TUNEL method demonstrated that cell apoptotic index in lung cancer tissues was significantly lower than that in lung tissues around tumor(P<0.05), the cell apoptotic index in small cell lung cancer tissues was respectively significantly lower than that in adencarcinoma and squamous carcinoma tissues,the expression of three groups had significantly difference(P<0.05). By correlation analysis the expression of NF-κB-p65mRNA was negative correlated with apoptotic index in lung cancer tissues.Conclusions1. The expression of NF-κB-p65mRNA and MMP-1 in lung cancer tissues was significantly higher than in lung tissues around tumor, and their expression was associated with TNM staging, tumor cell differentiation,the lymph node metastasis and pathological typing. By one-factor analysis of variance, the expression of NF-κB-p65mRNA was correlated with the expression of MMP-1 in lung cancer tissues. NF-κB may regulate the expression of MMP-1 by some means.2. The expression of NF-κB-p65mRNA and CD15s in lung cancer tissues was significantly higher than in lung tissues around tumor, and their expression was associated with tumor cell differentiation,the lymph node metastasis and pathological typing. By one-factor analysis of variance, the expression of NF-κB-p65mRNA was correlated with the expression of CD15s in lung cancer tissues. NF-κB and CD15s may play a stimulative role in carcinogenesis and tumor cell metastasis and infiltration3. Apoptotic index in lung cancer tissues was significantly lower than that of cell in lung tissues around tumor, by correlation analysis the expression of NF-κB-p65mRNA was negative correlated with apoptotic index in lung cancer tissues.NF-κB may participate in apoptotic process of lung cancer tissues.
