Studies On Bio-active Constituents Of Belamcanda Chinensis And Eleutherine Americana

In this paper,we studied on the preparation of total isoflavones of Belamcanda chinensis, and the biologically active compounds from Belamcanda chinensis and Eleutherine americana.The Traditional Chinese Medicine She-gan(Belamcanda chinensis) has been used as folk medicine for the treatment of coughing and so on. As a common folk medicine,it has been planted in many provinces,such as Anhui,Hubei,Yunnai,Sichuan and so on.To ensure the prepartion technology of total isoflavones in Belamcanda chinensis,we studied the chenical constituents of the rhizomes of Belamcanda chinensis.The dried rhizomes were chopped and extracted with 80% ETOH three times under reflux.The ETOH extract was concentrated and filterd.Put for 12h,and we get filtrate and precipitation.The content of total isoflavones was determinated by UV and HPLC.50g extract of ETOAC and ETOH were isolated by silica gel,Sephadex LH-20,and octadecylsiiyl gel,13 compounds were isolated and 11 of them were identified on the basis of chemical reaction and spectral analysis.They are:tectoridin(Ⅰ), acetovanillone(Ⅱ), 4-hydroxy-acetophenone(Ⅲ),β-sitosterol(Ⅳ),β-daucosterol(Ⅴ),5,7,4'-trihydroxyl-3',5'-di methoxyflavone(Ⅵ),luteolin(Ⅶ),apigenin(Ⅷ),5,7,4'-trihydroxyflavanones(IX),isorharnne tin(Ⅹ).CompoundsⅥ,Ⅶ,Ⅷ,Ⅸwere isolated for the first time from this plant.The content of total isoflavones of Belamcanda chinensis was determinated,and the measures were also investigated.The result indicate that the measures was right and the content of isoflavones is above 70%.Eleutherine americana,a Chinese traditional medicine,have been used for the treatment of arthralgia,dizziness and so on.It is a widely growing plant species in Yunnan,Guangxi province.The dried bulbs of Eleutherine americana were chopped and immersed with 80% ETOH,one werk every time and total three times. The ETOH extract was concentrated and filterd. Put the extraction into water and extracted with Petroleum ether,Etoac and Butanol.The extraction of Petroleum ether and Etoac were isolated by silica gel,Seph-adex LH-20,and octadecylsiiyl gel,23 compounds were isolated and 16 of them were identified on the basis of chemical reaction and spectral analysis.They are:(coumaric acid Ea-1),(vanillin Ea-2), (ethyl hydroxybenzoate Ea-3), (ferulaldehyde Ea-4), (3-methoxy-ethyl-4-hydroxybenzoate Ea-5), (8-hydroxy-3,4-dimethoxy--1-methyl anthraquinone-2-carboxylic acid methyl ester Ea-6), (4,8-dihydroxy-3-methoxy-1-methyl-anthraquinone-2-carboxylic acidmethyl ester Ea-7),ω-Hydroxypropioguaiaco-ne(Ea-8), (ferulate Ea-9), (stiosterone Ea-10), (eleutherin Ea-11), (isoeleutherin Ea-12), (3-methoxy-4-hydroxy-propyl-alcohol Ea-13),(eleutherol Ea-14), (stigmas-terol Ea-15), (β-sitosterol Ea-16).
