Objective:Observing the effects of model on protein expressions of related factors in the mitochondrial pathway of the articular cartilage in rats with exercise-induced arthritis and the serological changes. Ibuprofen as positive control, investigate the possible molecular mechanism for exercise-induced arthritis and the mechanism of Huwentoxin-I (HWTX-I)on protecting exercise-induced arthritis is researched, so as to provided theoretical and experimental basis for development and progression of the exercise-induced arthritis.Methods:96 grown-up male SD rats, which were randomly divided into the early physiological saline group,early HWTX-I administered group and early Ibuprofen administered group, late physiological saline group,late HWTX-â… administered group and late Ibuprofen administered group, adopting the model of the improved Hulth model combined with intermittent low-intensity exercise, adopting Yaksh T. L, Rudy T. A catheterization in spinal epidural space in the study, relative drugs were given to rats through the reserved PE10 plastic tubes or intragastric administration. Observing the effects of HWTX-I on morphological change of articular cartilage with microscope, the protein expressions of Bcl-2,Bax,Caspase-3,Caspase-9 in mitochondrial apoptosis pathway in the articular cartilage of the rats with exercise-induced arthritis with immunohistochemistry, spectrophotometry was used to detect the activity of SOD,CAT,GSH-Px and the concent of NO in serum.Results:(1) By HE staining, in early physiological saline group, the tide mark became disappeared in knee articular cartilage, chondrocyte ranged disorderly, some chondrocyte underwent pyknosis;in early Ibuprofen administered group, the tide mark became unclear, chondrocyte ranged nou-uniformity, very less chondrocyte underwent pyknosis;in early HWTX-â… administered group, approximating to that of normal cartilage. In late physiological saline group, articular cartilage defect the tide mark became disappeared, chondrocyte ranged disorderly; in late Ibuprofen administered group, the tide mark became unclear, chondrocyte ranged nou-uniformity, very less chondrocyte underwent pyknosis; in late HWTX-â… administered group, approximating to that of normal cartilage, the tide mark orderly, chondrocyte ranged nou-uniformity, some chondrocyte underwent pyknosis.(2) By immunohistochemistry staining, there were the strongest Bcl-2 protein expressions in the knee articular cartilage of rats in the HWTX-â… administered group (yellow represented the positive expression of proteins); followed by the Ibuprofen administered group;in the physiological saline group, strong expressions of each factor protein could be observed, but there were fewer positive cells than the HWTX-â… administered group and the Ibuprofen administered group. There were the strongest Bax,Caspase-3 and Caspase-9 protein expressions in the knee articular cartilage of rats in the physiological saline group;followed by the Ibuprofen administered group; in the HWTX-I administered group, strong expressions of each factor protein could be observed, but there were fewer positive cells than the physiological saline group and the Ibuprofen administered group.(4) By spectrophotometry detection, the activity of SOD,GSH-Px were the highest in the HWTX-I administered group, followed by the Ibuprofen administered group, and the lowest in the the physiological saline group;the activity of CAT the lowest in the the physiological saline group; the concent of NO in serum the highest in the the physiological saline group, followed by the Ibuprofen administered group, and the lowest in the the HWTX-I administered group.Conclusion:(1) The administration of HWTX-â… in spinal dura mater space up the protein expressions of Bcl-2 in mitochondrial apoptosis pathway and down the protein expressions of Bax,Caspase-3 and Caspase-9, which has obviously protective effects on rats articular cartilage damage induced by exercise-induced arthritis.(2) The administration of HWTX-I in spinal dura mater space up the activity of SOD,CAT,GSH-Px and down the concent of NO in serum, which can obviously relieve the damager free radical in the exercise-induced arthritis rats'articular cartilage(3) Mitochondrial pathway is closely correlated with exercise-induced arthritis. It is of important theoretical significance to explain the mechanism of mitochondrial pathway in the exercise-induced arthritis.(4) The movement of calcium has high correlated with the development of exercise-induced arthritis. Our research has approached the molecular mechanism of exercise-induced arthritis and the efficacy of calcium channel blocker, it is significance for prevention and cure of exercise-induced arthritis.
