Purpose:Sijunzi Tang, Shen Ling Baizhu San, Buzhong Yiqi Tang are famous agent of strengthening spleen and replenishing qi in the Chinese Tranditional Medicine. Because the three prescription include different Chinese herbs, on the basis of strengthening spleen and replenishing qi, there were nourishing gastric, invigorating spleen to resolve dampness and checking diarrhea, benefiting Qi and raising Yang respectively.At the same time, the three prescriptions have their different suitable gynecologic disease. If the three prescriptions act on the same animal modle with spleen deficiency syndrome, whether there will exist different sphere of action and effect degree and have selective effect on different pathological and physiological aspects. There are no reports of the effect of similarities and differences about the three strengthening spleen and replenishing qi prescriptions on the basis of "the correlation between fomula and sydrome".The study based on "the correlation between fomula and sydrome", from strengthening spleen and replenishing qi associated prescriptions act stating point. Adopted by excess fatigue+improper diet to form the modle. On the basis ofMethod:The male wistar rats were randomly assigned into the normal group (group 1), the spleen-deficiency group. The models were established by excess fatigue plus out of constant diet method for 4 weeks. On the 15th day, the models rats were randomly assigned into the the spleen-deficiency group (group 2), Buzhong Yiqi Tang gropu (group 3), Si Junzi Tang group (group 4), Shenling Baizhu San group (group 5). Rats of group 3,4 and 5 had been respectively fed with medicine of Buzhong Yiqi Tang, Si Junzi Tang,Shenling Baizhu San 4.05g-kg"1 for two weeks. The ones of the group 1 and group 2 were fed with equivalent physiological saline for two weeks. Observed and recorded all of the diet in rats, appearance of urine, hair color, active state in rats, and so on. Measured the body weight, rectal temperature, grip strength two times every week. On the 28th day, all rats 16 hours after fasting at the dose of 10% D-xylose solution 5ml per mouse. On the 29th day, the 10%of chloral hydrate anesthesia animals, separation test Plasma and Serum, skeletal muscle, place the skeletal muscle into the 10% formalin, clipping subparagraph small intestine, colon, complete kidney, spleen, liver. Putting parts of the tissues, separated test Plasma and Serum into the vials and placing them into-80 degrees Celsius freezer. Using radio immunity and enzyme linked immunoassay detect all indexes. Parts of skeletal muscle in the 10% formalin would be harvested foe HE sections and observed pathological state in the light microscope. Statistical methods:the data were described by (x±SD) and the number of samples compared between groups used one-way ANOVA (one-way ANOVA) test:non-normal data using non-parametric statistical analysis. All data used SPSS 20.0 statistics software.Results:1. The appearance representations, the body weight, rectal temperature, grip strength, nerve function, nervous-endocrine-immune network, water-electrolyte metabolism and water transport, the changes of skeletal muscle power of rats model with spleen-deficiency.(1) During the experiment, the normal group rats had no obvious change. On the 15 th day, the models rats shown the abnormal changes of behavior, less activity, even get together,manifesting lassitude, drowsy, withered hairs, stool loosed, the diet of the model rats decreased, and so on. (2) During the experiment, the body weight of the normal group rats increased gradually, the rectal temperature changed little, grip strength increased gradually with the increase of body weight. Compared with normal group, the model rats of the body weight, the rectal temperature, the grip strength decreased significantly(p<0.01). (3) the model rats of plasma D-xylose, MTL, GAS and the serum P-EP, Ach, T3, T4, IL-2, IL-6, cAMP, cAMP/cGMP decreased significantly, the plasma of SS and cGMP increased significantly (P<0.05, P<0.01). (4) Compared with normal group, the model rats plasma levels of ADH, ALD and serum Na+, K+, kidney AQP2 content increased significantly, small intestine and colon mucosa AQP3 decreased significantly (p<0.01). (5) Compared with normal group, the model rats skeletal muscle levels of grip strength, SDH, CK-MM, Ca2+-ATPase, M-AChRRM2, Musclin decreased significantly, serum AchE increased significantly (p<0.01,p<0.05). The HE shown muscle fiber disordered, uneven thickness, gap widened. Vacuoles were formed inside the muscle fibers,muscle atrophy,inflammatory cell infiltration and phagocytosis.2. Effects of the three different prescriptions on the changes. (1) Compared with model group, the behavior of integral value of the three medicated groups had improvement to some degrees, the body of weight and body temperature all increased significantly (p<0.05). (2) Compared with model group of plasma D-xylose, MTL, GAS and the serum (3-EP, Ach, T3, T4, IL-2, IL-6, cAMP, cAMP/cGMP increased significantly, the plasma of SS and cGMP decreased significantly (P<0.05, P<0.01). (3) Compared with model group rats of the plasma levels of ADH, ALD and serum Na+, K+, kidney AQP2 content decreased significantly, small intestine and colon mucosa AQP3 increased significantly (p<0.01). (4) Compared with model group, three medicated groups of skeletal muscle levels of grip strength, SDH, CK-MM, Ca2+-ATPase, M-AChRM2, Musclin inreased significantly, serum AchE decreased significantly (p<0.01,p<0.05).The HE shown muscle fiber ordered, even thickness, gap thinner. Vacuoles disappeared no,inflammatory cell infiltration and phagocytosis.3. Effects of the three different prescriptions on the all indexes of difference changes. (1) On the appearance representations and nerve function of rats model with spleen-deficiency, the integral value of active state, skin and hair, total integral value, the rectal temperature and grip strength in bu zhong yi qi tang group improved more significantly than the other two prescriptions. (p<0.01, p<0.05). (2) Compared with group 4 and 5, there were differences of the increased IL-6, cAMP cAMP/cGMP and decreased cGMP in group 3 (p<0.01,p<0.05). Compared with group 5, there were differences of the increased GAS and β-EP in group 3 (p<0.01,p<0.05), had an increased tendency from group 4. The serum T3 and Ach improved better in the group 5 from the other prescriptions. (p<0.05). (3) Compared with group 3 and 4, the plasma levels of ADH, ALD and serum Na+, K+, kidney AQP2 content decreased significantly, small intestine and colon mucosa AQP3 increased significantly in group 5(p<0.01, p<0.05). Compare with group 4, the kidney AQP2 content decreased significantly in group3 and 5 (p<0.01,p<0.05).(4) Compared with group 4 and 5, skeletal muscle levels of grip strength, SDH, CK-MM, Ca2+-ATPase, M-AChRM2 increased significantly in group 3(p<0.01, p<0.05).Conclusion:The models with spleen-deficiency were established by excess fatigue plus out of constant diet method for 4 weeks. Not only the models rats exit similar to clinical patients spleen symptoms in appearance, but also represent characteristic to change in appearance, all exist abnormal function of the digestive system, immune system, water and salt metabolism, autonomic function and skeletal muscle system. Three different reinforcing spleen recipes on the spleen-deficiency symptoms, temperature, vegetative nerve function and multiple system of rats model with spleen-deficiency syndrome had marked improvement, however, to improve different type of symptoms with particular focuses respectively. Compared with the base reinforcing spleen recipes si jun zi tang, the bu zhong yi qi tang had effect and gives prominence on body temperature and skeletal muscle strength, shen ling bai zhu san had prominent role on the water and salt metabolism and water transport.Spleen-deficiency syndrome is a pathological and physical state with multiple system abnormalities, the effects of different reinforcing spleen recipes are similar in different and particular focuses respectively on the pharmacological effects.
