Effects Of Acupuncture At "Neiguan" On The Expression Of CLC-3 And CLC-4 Proteins In Myocardial Tissue Of Myocardial Ischemia Mice

Purpose:To investigated and compared the impacts of acupuncture on the expressions of cardiac chloride channel CLC-3 and CLC-4 protein of the ASIC3 knock out mice(ASIC3^-/-) with myocardial ischemia via peritoneal injection of isoproterenol（ISO） and to discuss the mechanism of along-channel acupuncture,which would provide experimental evidence of acupuncture prevention myocardial ischemic for clinical applications.Material and method:37 SPF ASIC3^-/- mice were randomly divided into 6 groups : Blank group（7）, modeling control group（6）, Neiguan treatment group（6）, Lieque control group（6）, Zusanli control group（6） and Non-acupoint control group（6）, and make them myocardial ischemia by subcutaneously injection of isoproterenol（ISO）, except the blank group. Electroacupuncture treatment was given to Neiguan（PC6） treatment group, Lieque（LU7） control group, Zusanli（ST36） control group and Non-acupoints control group once a day, 20 minutes each time at 8:00-11:00 daily and treatment for 7 days, while the blank group without acupuancture and the modeling control group is also the same after modeling. Then cardiac tissue were observed by microscope after HE staining.The content of MDA was detected by Colorimetric method, protein expression of CLC-3 and CLC-4 were detected by Western blot method.Results:1. Compared with blank group, the Lieque（LU7） control group, the Zusanli（ST36） control group and the nonacupoint control group exist various ischemic area of cardiac tissue which was observed by HE staining; Compared with modeling control group, the Neiguan（PC6） group can relieve the situation of myocardial ischemia Significantly.2. Compared with the blank group, the serum MDA levels of the modeling control group, the Lieque（LU7） control group, the Neiguan（PC6）treatment group,the Zusanli（ST36） control group and the non-acupoint control group were significantly ingcrease（p<0.01）; But the MDA levels are clearly reduced in Nei Guan（PC6） group（p< 0.01） against the one in modeling group, so the same in the Zusanli（ST36）group（p<0.05）, No differences were found among the modeling control group, the Lieque（LU7） control group, and the nonacupoint control group（p>0.05）; Compared with Nei Guan（ST36） group, MDA levels are clearly higher in the Lieque（LU7） control group, and the nonacupoint control group.4. Compared with blank group,there is a significant increased in the contents of CLC-3 and CIC-4 CL- channels protein in the modeling group,the Lieque（LU7） group, the Zusanli（ST36） group and the Nonacupoint group（p<0.01）; But the contents of CLC-3 and CIC-4 CLchannels protein are reduced in Nei Guan（PC6） group and Zusanli（ST36） group against the one in modeling group, No differences were found among the modeling control group and the Lieque（LU7） control group, and the nonacupoint control group（p>0.05）; Compared with Nei Guan（PC6） group, the contents of CLC-3 and CIC-4 CL- channels protein are clearly higher in the Lieque（LU7） control group, and the nonacupoint control group.Conclusion:1.Along-channel acupuncture at Neiguan（PC6） acupoint can effectively relive the pathological conditions of myocardial ischemia via regulating the contents of CLC-3 and CIC-4 CL- channels protein.2. Acupuncture at Neiguan（PC6） and Zusanli（ST36） point can both regulating the myocardial ischemia,but the therapeutic effects of Neiguan（PC6） group is better than Zusanli（ST36） group via comprehensive comparison among changes of each indicator.Therefore, along-channel acupuncture the Neiguan（PC6） to treatment of myocardial ischemia is specificity.
