Objective:To evaluate the protective effect of acupuncturing Neiguan on myocardial ischemia rats,and to explore the mechanism of myocardial purine signaling in rats with myocardial ischemia.Methods:SD rats were selected as the research object,which were divided into blank group,sham operation group,model group,Neiguan group,Hegu group,non-acupoint group.The model of myocardial ischemia was established by ligating the left anterior descending coronary artery.The rats in the Neiguan group,hegu group,non-acupoint group were given PC6,L14,non-acupoint electroacupuncture treatment after second days,once a day for 5 days.Blank group,sham operation group,model group rats were only bundled and not given electroacupuncture treatment.The physiological recorder,TTC staining,HE staining and TUNEL were used to detect ECG,myocardial infarct size,myocardial tissue morphological changes andapoptosisofmyocardialcellnumberandmorphology;andthe immunohistochemistry and HPLC method were used to detection of ARA1,ARA2a,ARA2b,AR A3 and the content of ATP,ADP,AMP,ADO.Results:1.Changes of ECG in rats:model group,Neiguan group,Hegu group,non-acupoint group rats ST elevation,T wave height after operation,indicating that the model was successful.Sampling before each ST segment of rat voltage comparison:compared with the model group,Neiguan group,Hegu group,non-acupoint group ST voltage rised?P<0.05?.2.Changes of myocardial infarction area in rats:compared with the model group,the proportion of myocardial infarction area in Neiguan group,Hegu group,non-acupoint group decreased?P<0.05?.3.The morphological changes of myocardial tissue of rats:in blank group and sham operation group,the arrangement of myocardial fibers was close and the morphology of myocardial cells was intact.The myocardial cell in the model group was disordered,the ventricular wall was thin and the focal connective tissue hyperplasia and fibrosis.The cell in Neiguan group was disordered,and the muscle fibers were loose and edema.In Hegu group and non-acupoint group,myocardial rat focal connective tissue hyperplasia.4.Changes of myocardial apoptotic cells in rats:compared with model group,the apoptotic rate of myocardial cell in Neiguan group,Hegu group,non-acupoint group were decreased?P<0.01?.Compared with Neiguan group,the apoptotic rate of myocardial cell in Hegu group,non-acupoint group were increased?P<0.01?.5.Changes of purine content in rats myocardium:compared with the blank group,the content of ATP in the model group decreased?P<0.05?,the content of ADP,AMP and ADO in myocardium increased?P<0.05?.Compared with the model group,the content of ATP in myocardium increased?P<0.05?in Neiguan group,the content of ADP and AMP in the Neiguan group,Hegu group,non-acupoint group decreased?P<0.05?.6.Changes of rats myocardial adenosine receptor expression:compared with the blank group,the average optical density of ARA1,A2aa and A2bb increased?P<0.05?in the model group.Compared with the model group,the average optical density of ARA1,A3 decreased?P<0.05?in Neiguan group,and the average optical density of ARA2aa increased?P<0.05?in Hegu group and non-acupoint group,and the average optical density of ARA2bb increased?P<0.05?in Hegu group,and the average optical density of ARA3 increased?P<0.05?in non-acupoint group.Conclusion:1.Acupuncture Neiguan has protective effect on myocardial ischemia in rats,it can be achieved by narrowing the area of myocardial infarction,improving the morphological structure of myocardium and reducing the apoptosis of myocardial cells to protect myocardium.3.Acupuncture Neiguan to improve the mechanism of purine signaling in rats with myocardial ischemia may be:inhibition of ATP consumption,slow ATP decomposed into ADP,AMP,ADO;while inhibiting of adenosine,and down-regulation of ARA1and A3 expression,up-regulation of ARA2aa expression.
