Abstract:Study of Xiaoyao Pill on liver stagnation and spleen deficiency type non intervention effect of expression of Nonalcoholic fatty hepatitis in rat liver TLR4/TRIFMethods: 32 male SD rats were randomly divided into four groups:the control group, model group, Xiaoyao San group and glycine group, 8 rats in each group. The control group was fed with basal diet, the other groups were treated with high glucose and high fat diet and diet, chronic restraint stress stimulation for 14 weeks.Spearheading the group, glycine group were given on the basis of modeling spearheading the lysate,glycine gavage,1 times/d, for 30 days,abdominal aortic blood, serum total cholesterol(TC), triglycerides(TG), high density lipoprotein(HDL-C), Low density lipoprotein(LDL-C), transaminase(ALT), Free fatty acids(FFA),Tumor necrosis factor(TNF-α), norepinephrine(NE),5-hydroxyl tryptamine(5-HT), D-xylose excretion rate,Fasting blood glucose(GLU),Fasting insulin(FINS), Insulin resistance index(FIRI); HE staining of liver morphology;Western blotting technique to detect the expression of TLR4 and TRIF.Results:The serum index in model group was compared with that in control group:the model group TC、TG、ALT、FFA、LDL-C、TNF-a、LPS、GLU、FINS、FIRI levels were significantly increased, HDL-C levels were significantly lower; The neurotransmitter in model group was ompared with that in control group:5-HT, NE,Compared with control group: the model group D-xylose excretion was significantly reduced; fat vacuoles in liver cells cytoplasm, significantly increased the expression of TLR4 and TRIF in liver; compared with the model group, the treatment group TC、TG、ALT、FFA、LDL-C、TNF-α、LPS、GLU、FINS、FIRI decreased, HDL-C, 5-HT, NE, D-xylose excretion rate increased protein expression of TLR4 and TRIF reduced accordingly changes consistent treatment group.Conclusion: Liver spleen syndrome Nonalcoholic fatty hepatitis model to replicate the success, in line with liver spleen syndromes, TLR4 and TRIF expression spearheading reduce. |
