The Cold-pressed Sesame oil was studied in this paper,and Sesame oil for injectionwas prepared by acid degumming, alkali refining, bleaching and deodorizationprocess. The Cold-pressed Sesame oil basic physical and chemical indicators wereexamined.The changes of phospholipid content of Sesame oil, peroxide value andacid value during acid degumming, acid value during alkali refining process werestudied. And the impacts of different adsorbents on absorbance of sesame oil werecontrasted. On this basis, the effect of deodorizing treatment on sesame oil qualitywas studied. The main conclusions were as follows:1.The physical and chemical indicators of crude oil were: absorbance (450nm)0.214,phospholipid content199.61mg/kg, acid value1.88mgKOH/g, the peroxide value4.19mmol/kg, unsaponifiable matter0.15%, saponification value186mgKOH/g.The indicators didn’t meet the requirements of the pharmaceutical oil.2.Degumming process of Cold-pressed Sesame oil was studied. The best conditions ofacid degumming were obtained by orthogonal experiment: temperature60℃,citricacid dosage5.0g/kg, water dosage2.5%,time40min.Under the conditions ofdegumming rate was72.9%. Degummed Sesame oil ’s color obviously shallowed,Unsaponifiable matter content and AV lowed, POV rised.3.Deacidification process of degummed Sesame was studied. The best conditions ofalkali deacidification were obtained by orthogonal experiment:alkali concentration8Bé, excessive alkalia quantity0.1%, temperature70℃, time20min.Under theconditions, the refining rate was90.8%, acid value0.06mgKOH/g, and acid valuecould meet requirement of oil for injection (0.1mgKOH/g) in "ChinesePharmacopoeia".4.Adsorption bleaching processes of sesame oil was studied.Firstly, adsorbents werescreened and selected activated clay and diatomaceous earth as hybrid sorbent. Thebleaching process was optimized by response surface and the best conditions asfollows: mixing ratio of diatomaceous earth and activated clay1:4, bleachingtemperature91℃, bleaching time21min, bleaching dosage3.7%. Under the bestconditions, absorbance (450nm) of sesame oil0.039, bleaching rate reached78.65%.Except for the clay odour of bleaching Sesame oil, other quality indexes reached thestandards of oil for injection.5.Isotherm of absorbent bleaching process of activated clay and diatomaceous earth was studied. The bleaching process fitted the Freundlich adsorption equation,Freundlich model could be used to evaluate the performance of the adsorbent andguide practice.6.Deodorizing process of Sesame oil was studied. Deodorizing conditions were asfollows: deodorizing temperature180℃, time30min, vacuum0.099MPa.The claysmell of Sesame oil could be removed by deodorizing.Quality indexes of deodorized Sesame oil were: Iodine value111g/100g,saponification value of194mgKOH/g, acid value0.08mgKOH/g, peroxide value1.08mmol/kg, unsaponifiable matter0.56%and absorbance (450nm)0.035.Qualityindexes of Sesame oil reached requirements of oil for injection. Sesamin andSesamolin in Sesame oil were0.3470%and0.1462%respectively.The results indicated that refining process of edible oil refining was suitable for oilfor injection. Sesame oil for injection was prepared by acid degumming, alkalinedeacidification, adsorption bleaching and deodorization process is feasible.
