Inula helenium L.(Compositae) is a perennial plant, distributed in many areas ofChina, Europe, and North America. The root of I. helenium is rich in eudesmane-typesesquiterpene lactones, such as isoalantolcatone and alantolactone. In recent years,extensive investigations on active constituents of I. helenium roots proved thatsesquiterpene lactones are responsible for various biological activities, such asantibacterial, antifungal, hepatoprotective and antitumor activity. Therefore, it isnecessary to find an efficient method of extraction and purification of sesquiterpenelactones.The current study was mainly focused on the ultrasound-assisted extraction ofphenolic and microwave–assisted extraction of flavones. The tests by orthogonalarray design [L9(34)] were performed to analyze the effect of single factors on theyield such as ethanol concentration, extraction time and times. As the result, theoptimal extraction conditions were found as follows: the dried roots of I. heleniumwere extracted by reflux with95%ethanol in materiel/liquid ratio of1:10(w/v) for2hours to produce the ethanol extract. The optimized purification of ethanol extractwas achieved by a pressurized silica gel (100-200mesh) column chromatographywhose ratio of diameter to high was1:2. The ratio of mixed sample to silica gel was1:3(w/w), the eluent, composed of petroleum ether (30-60℃) and acetone (100:3,v/v), flowed at the rate of35ml/min. The extraction rate of the lactones obtainedusing the optimal method was above70%, and the purity was above90%. Then thequality standard was proposed.
