Juglans mandshurica Maxim. and Cortex Dictamni. as traditional herbal medicine,contains a variety of natural active ingredients.Both have many pharmacologically activity of anti-bacterial,anti-oxidation, anti-inflammatory,antiviral,et al.In recent years, studies have shown Juglans mandshurica Maxim. and Cortex Dictamni.also have insecticidal activities, however, study on the insecticidal activities than less. Therefore, study on the extraction,separation,structural identification of active components and insecticidal activities in Juglans mandshurica Maxim. and Cortex Dictamni.have important significance for environmental protection.First, Juglans mandshurica Maxim. and Cortex Dictamni.were to optimize the extraction process.The optimum extraction conditions of juglone from Juglandis Mandshuricae Cortex was solid-liquid ratio of 1:12, the ethanol concentration of 95%, was extracted three times, each time the reflux of 50 min. The optimum extraction conditions of juglone from Juglans mandshurica Maxim.leaf was solid-liquid ratio of 1:12, the ethanol concentration of 75%, was extracted three times, each time the reflux of 50 min.And the content of juglone from Juglans mandshurica Maxim more than the content of juglone from Juglandis Mandshuricae Cortex. The optimum extraction conditions of Cortex Dictamni.was solid-liquid ratio of 1:7, the ethanol concentration of 75%,choose ASE extraction three times at 70 ℃, each time the reflux of 4 min.In the present study,the solvent systems consisted of n-hexane-ethylacetatemethanol-water- butanol(0.8:1:1:1:0.16,v/v/v/v/v)for HSCCC was choose,the revolution speed was 850 rpm/min,the flow rate was 2.0 m L/min,the separation temperature was 25 o C,and the detection wavelength wan 228 nm.Four single compounds were obtained from 200 mg of ethanol extract of Cortex Dictamni. in one-step separation by HSCCC, the four single compounds were limonin, dictamnine,obacunone and fraxinell- one according to the information of the ion fragmentation by HPLC-MS.Last, study on the insecticidal activities in ethanol extract of Juglans mandshurica Maxim. and Cortex Dictamni..The test was performed by immersion method for ethanol extract of Juglans mandshurica Maxim.leaf. The results showed that extracts from Juglans mandshurica Maxim.leaf had a stomach action on Trialeurodes vaporariorum,had contact toxicity on Plutella xylostella L. and Acythosiphon pisum. The test was performed by drip methodand for ethanol extract of Cortex Dictamni. The results showed that ethanol extract of Cortex Dictamni., dictamnine and fraxinellone had contact toxicity on Mythimna separata、Monolepta hieroglyphica,Callosobruchus chinensis,no contact toxicity on Henosepilachna vigintioctopunctata and Heliothisdipsacea; ethanol extract of Cortex Dictamni and fraxinellone had stomach action on Ambrostoma quadriimpressum Motschlsky; ethanol extract of Cortex Dictamni. had contact toxicity on Pierisrapae Linne;obacunone no insecticidal activities.
