Juglans mandshurica is an important medicinal plant in China,the green peel has anticancer,antibacterial,antioxidation,anti-virus and other functions,so the research and development of the green peel of Juglans mandshurica has both objective economic prospects and good social benefits.This paper studied the extraction and separation of active substances from the the green peel of Juglans mandshurica and optimized the extraction conditions,to provide experimental data and theoretical basis for the development and utilization of iuglans mandshurica resources.The active substances of the green peel of Juglans mandshurica were extracted by 60%ethanol,and petroleum ether,chloroform and ethyl acetate were used to extract the active substances.After separation by chromatography,the active ingredients were detected and analyzed by gas chromatography and the GC-MS.Petroleum ether was selected as the eluent in the extraction phase of petroleum ether:ethyl acetate=90:10 mixed solution,the separation results contain six effective substances;In the chloroform extraction phase,the eluents were petroleum ether:ethyl acetate=70:30 and petroleum ether:ethyl acetate=40:60.The separation results contain 8 kinds of effective substances and 6 kinds of effective substances;The eluent of ethyl acetate extraction phase was chloroform:methanol=90:10 mixed solution,and the separation results contain 9 effective substances.On the basis of single factor,response surface methodology was used to optimize the extraction process of total anthraquinones from green peel of Juglans mandshurica,and the optimum extraction conditions were explored.The optimum extraction conditions of total anthraquinones from leaves of Juglans mandshurica were studied by the same method,and compare their similarities and differences.The results obtained by response surface methodology showed that the order of influence of each factor on the extraction rate of Total anthraquinone was as follows:Liquid-solid ratio(A)>ethanol concentration(B)>ultrasonic temperature(C)>ultrasonic time(D).The optimum extraction conditions of total anthraquinones from green peel of Juglans mandshurica were as follows:liquid-to-material ratio 15:1,ethanol concentration 75%,ultrasonic temperature 45?,ultrasonic time 45 min,under these conditions,the extraction rate of total anthraquinone was 3.25 mg·g-1.The optimum extraction conditions of total anthraquinone from leaves of Juglans mandshurica were as follows:liquid-to-material ratio 15:1,ethanol concentration 80%,ultrasonic temperature 50?,ultrasonic time 50 min;under these conditions,the extraction rate of total anthraquinone was 2.10 mg·g-1.Study on antioxidant activity of total anthraquinones extracted from green peel of Juglans mandshurica,the results show that anthraquinones in green peel showed significant scavenging effects on hydroxyl radicals,nitrite ions,superoxide anions and DPPH radicals,the scavenging ability of total anthraquinone to four free radicals was DPPH free radical>hydroxyl free radical>superoxide anion>nitrite ion.The scavenging capacity of anthraquinones to four free radicals was stronger than that of ascorbic acid,which indicated that anthraquinones from green peel of Juglans mandshurica had strong antioxidant activity and were a good natural antioxidant.The total anthraquinones from green peel of Juglans mandshurica under the optimum extraction conditions were studied by perforation method.The results showed that the above substances could inhibit the growth of both bacteria and had obvious bacteriostasis effect.
