The active components of the Juglans mandshurica leaves was extracted with ethanol,and was separated by petroleum ether,chloroform,ethyl acetate and acetone.The extracted phases were chromatographically separated and analyzed by gas chromatography(GC)and gas chromatography-mass spectrometry(GC-MS)to identify the active components and structures.The petroleum ether extraction phase of petroleum ether-ethyl acetate system was divided into five components when the ratio of 95:5.And the chloroform extraction phase of chloroform-methanol system was divided into three components when the ratio of 89:11.While three parts of the ethyl acetate extraction phase can be separated at the ratio is 76:24,and two components at ratio is 70:30 of methanol-chloroform system.The fraction of the acetone-methanol system of the acetone extraction phase separated two components when the ratio is 92:8.The single factor and response surface methodology was used to optimize the extraction conditions of polyphenols from Juglans mandshurica,and determine the optimal extraction conditions for polyphenols in leaves.At the same time,compared with the green peels.The optimal conditions for polyphenols in leaves as follows:the volume fraction of ethanol was 70%,and the ratio of liquid to the material was 15:1,the ultrasonic extraction time was 30 min,the ultrasonic extraction temperature was 60?.As for the green peels,the optimal conditions as follows:the volume fraction of ethanol was 60%,and the ratio of liquid to the material was 25:1,the ultrasonic extraction time was 40 min,the ultrasonic extraction temperature was 60?.The maximum extraction rate of polyphenols in leaves was 3.46%,and As for the green peels,the maximum extraction rate of polyphenols was 1.37%.The biological activity identification was mainly conducted from two aspects of antioxidant activity and antibacterial activity.Four types of free radicals were used to test the antioxidant capacity of polyphenols.The test results prove that the scavenging of DPPH free radicals by the polyphenols in the Juglans mandshurica leaves and green peels was greater than the superoxide anion and was also greater than the hydroxyl radical,and the scavenging effect on the nitrite ion was the weakest.The antibacterial activities of the polyphenols,the petroleum ether phase of the ethanol extract and the chloroform phase were analyzed using two strains of gloeophyllum trabeum and coriolus versicolor.It has certain antibacterial activity against gloeophyllum trabeum and coriolus versicolor.
