Ethanol-water mixed solution extract the powder of green peel of Juglans mandshurica by ultrasonic assisted extraction method, juglone content was determined by UV spectrophotometry. two substances which detected by GC-MS from the extraction of juglone in green peel of Juglans mandshurica with 40% ethanol as extraction agent are juglone content (84.84%) and (1R)-2,2-bismethyl-3-methylene-bicyclo[2.2.1]heptane(9.32%), impurities introduced by chloroform extraction solvent account for 5.84%. The juglone extract was separated and purified by column chromatography, orange-red needle-like crystals can be obtained through concentrate and the evaporation of solvent. identified as high purity juglone by using high performance liquid chromatography and nuclear magnetic resonance detection.Tannins was extracted by ultrasonic method from the green peel and leaves of Juglans mandshurica respectively,Tannins content was determined by UV spectrophotometry and phosphomolybdic acid - phosphotungstic acid assay (F-D method). Single variable method and the orthogonal experiment method were used to optimize tannin extraction technology respectively, meanwhile, the best extraction conditions were determined. The optimal extraction conditions of Single variable method for the tannins in green peel of Juglans mandshurica were the volume fraction of acetone 40%, solid-liquid ratio of 1:20, ultrasonic temperature of 50 ℃, extraction time of 30min. ultrasonic power of 105 W; The optimal extraction conditions for the tannins in leaves of Juglans mandshurica were the volume the fraction of acetone of 40%, solid-liquid ratio of 1:20, ultrasonic temperature of 40 ℃, extraction time of 30 min, ultrasonic power of 90 W; We select the volume fraction of acetone, ultrasonic temperature, ultrasonic time, solid-liquid ratio as filter conditions, each factor choose three levels. Useing L9(34) orthogonal test method optimizes tannin extraction technology. The optimal extraction conditions for the tannins in green peel of Juglans mandshurica were the volume fraction of acetone 40%, ultrasonic temperature of 40 ℃, extraction time of 40 min, solid-liquid ratio of 1:15. The optimal extraction conditions for the tannins in leaves of Juglans mandshurica were the volume the fraction of acetone of 50%, ultrasonic temperature of 60 ℃, extraction time of 30 min, solid-liquid ratio of 1:20.Finally, tannin of antioxidant capacity and antimicrobial activity was examinated. The results demonstrate that tannins in both green peel and leaves had excellent scavenging capacity for nitrite ions, hydroxyl radicaland superoxide anion. Tannins the green peel of Juglans mandshurica have the strongest antioxidant capacity for nitrite ions, The tannins in leaves have the strongest antioxidant capacity for superoxide anion. The antioxidation of tannin in leaves is better than that of tannin inthe green peel. Tannins in both green peel and leaves had excellent antibacterial effect for Staphylococcus aureus and Escherichia coli. For tannin in the green peel, the minimum antibacterial concentration of Staphylococcus aureus and Escherichia coli is 1.61 mg/L and 0.81 mg/L respectively. While the minimum antibacterial concentration of Staphylococcus aureus and Escherichia coli has the same data which is 1.54 mg/L for tannin in leaves.
