The Eucommia In Pinoresinol Glucoside Geniposide Acid Extraction, Purification And Determination

The extraction. purification and analysis method of two biologicallyactive compositions -Pinoresinol Diglucoside(PDG) and GeniposidicAcid(GPA) in Eucommia (a precious Chinese medicine)were investigated:(l)Technological extraction process of PDG and GPA with organicsolvent was experimented. The extraction ratio of PDG was 92.48%when extracted with 60 vol% ethanol aqueous, twice at 60 , 60min eachtime; the extraction ratio of GPA was 92.57% when extracted with 1:12of 50 vol% ethanol aqueous, once at 90 癈, 60min and the stability ofGPA in the solvent was also discussed elementarily. (2)Technologicalprocess of purification was completed by chromatographic columnpacked with macroteticular resin. PDG was absorbed well by S resin andeluted by 30-40 vol% ethanol aqueous. The recovery ratio of PDG inthe process was 44.5%. On the other hand, GPA was adsorbed well by Aresin, the optimum adsorption amount by GPA adsorption -desorptionexperiment was 17.28 mg/ml(wet resin). Employed 200*26mm column,eluted with 15 vol% ethanol aqueous at the flow rate 1.5ml/mm, thedesorption ratio and the recovery ratio of GPA were 91.15%,55.42% respectively. (3) Effects of molibe phase.acid, et al on theanalysis of PDG and GPA by HPLC were discussed as well. The contentof PDG and GPA in Eucommia were determined by reversed HPLC andthe analysis condition of PDG was as follow: 26 vol% methanol aqueous,UV detector at 228nm. The experimental separating time was 25min, thecalibration curve was linear in the range of the value of 0.096-1.92 u g,r=0.9997,the recovery was 87.8-110%; The analysis condition of GPAwas as follow: methanol-water-acetic acid=12:87:l(v/v), UV detector at240nm, the experimental separating was 35min, the calibration curve waslinear in the range of the value of 0.33-3.26 u g, r=0.9996,the recoverywas 92-106%.
