Maxingshiganfangji, composed of Ephedra herb, Amygdala amara, Plaster and Glycyrrhiza., was rooted from《ShangHanLun》which was writed by Zhang Zhongjing. It was commonly used for relieving cough, anti-inflammatory, dispelling the phlegm and suppressing asthma and widely applied in the treatment of acute and chronic bronchitis. This prescription was used well for years, but its prep technology was not adaptive to the modernization of traditional Chinese drug. Based on the modernization of traditional Chinese drug, this experiment investigated the prep technology systematically , also used HPLC to establish its quality standard and at the same time studied pharmacodynamics initially in Maxingshiganfangji.With the content of effective constituent as index, the influencing factors were studied by the orthogonal design. The best extract technology was accepted, it was as follows:10 times of water, soaking 6.0 hour, 3 times extractions(1.5 hours per time). The clarifier was 70% ethanol and the supplemental agents were 1% tween-80 and 0.2% sodium benzoate. The result showed that the extraction ratio of effective constituent in Ephedra herb was 60% and the ratio of extractum was 22%.A proper quality standard was established by HPLC to assure the effect and safety of the medicine. The content of ephedrine in Maxingshigankoufuye had been determined on the mobile phases consisted of acetonitrile-0.02mol/L potassium dihydrogen phosphate (4:100) with the wavelength of detector 207nm. The content of codeine phosphate in Maxingshigankoufuye had also been examined on the mobile phases consisted of 0.03mol/L sodium acetate-methanol(70:30)with the wavelength of detector 238nm. The results showed that the content of ephedrine was 0.432mg/ml and codeine phosphate was 2.697μg/ml in Maxingshigankoufuye. And the TLC method had also been used to discriminate Glycyrrhiza with the developing solvent consisted of n-Butanol- Glacial acetic acid-Water(4:1:2). The other exam items were also determined according to the CHP(2005).The pharmacodynamics had been investigated in Maxingshigankoufuye initially. Through relieving cough test, anti-inflammatory test, eliminating phlegm test and suppressing asthma test , this experiment determined its pharmacodynamics.The article was established in the eighth kind new drug(Traditional Chinese Drug). The pharmacological studies of the new drug were accomplished.
