Nucleic acid is one of the important biomacromolecules. The analysis of nucleic acids plays a very important role in life science. Deoxyribonucleic Acid (DNA), which is the main inheritance substance in biology, has the important biological function. Single nucleotide polymorphism (SNP) is the most familiar form of gene mutation. The detection of specific sequence of DNA and SNP is greatly important for clinical diagnosis, analysis of food and medicine, etc. Thus, it is very necessary to develop the simple and rapid detection methods with high selectivity for DNA and SNP analysis.Recently, nanoparticles are attracting more and more interest in biochemistry analysis. Metal nanoparticle is widely used in protein and nucleic acid analysis for its spectral speciality and biological compatibility. Biological nanoparticle is used in signal sensor, drug delivering and analysis of biochemical materials for its biological activity.In this contribution, we used gold nanoparticle and allele-specific PCR to develop a colorimetric method for SNP differentiation. The proposed method was convenient, rapid and reliable. We also differentiated SNP site of human p53 gene exon 8 using the proposed method.Ferritin is one important kind of biological nanoparticles. In acidic medium, Ferritin can catalyze luminol-H2O2 chemiluminescence (CL) reaction and produce strong CL signal. Based on this phenomenon, a simple and sensitive method for Ferritin determination has been developed. There was a good linear relationship between the CL intensity and the concentration of Ferritin in the range of 0.5 ng/mL-10 ng/mL, and the detection limit (3σ) was 0.36 ng/mL. The proposed method offers a new approach for direct detection of Ferritin as labeled nanoparticles.
