Enzymolysis will have a bright future in Chinese medicine industry.Using enzymolysis to optimize the extraction technology of Panax ginseng, Ganoderma lucidum,Schisandra.Selected schisandrin extracting amount as the determine index, the best condition of enzymolysis extraction from Schisandra were as follows:the amount of enzyme was 1.1%,the enzymolysis temperature was 70℃,enzymolysis 105 mins, after that extract twice with water.In enzymolysis extraction of Panax ginseng,we select different extraction process between water extracting,alcohol extracting,cellulase enzymolysis,pectinase enzymolysis and mixture enzymolysis of ginsenoside Rgl content from Panax ginseng,and the ginsenoside Rgl of cellulase enzymolysis has a higher extraction rate.After that we consider the best condition of cellulase enzymolysis were as follows:enzymolysis 60 mins, cellulase amount was 1.4%, the enzymolysis temperature was 45℃, after that extract twice with water,every time 1h.Use the content of polysaccharide as index, we adopt single factor experiment and orthogonal design method to optimize Ganoderma lucidum extract technology by cellulase enzymolysis and ultrasound method,and the best condition were as follows:the amount of enzyme was 2.5%, enzymolysis 60 mins,the enzymolysis temperature was 50℃, ultrasonic power was 300W, ultrasonic time was 6 min,and the content of polysaccharide was 30.17μg·g-1Use salvianolic acid as index, we adopt orthogonal design method to optimize Salvia extract technology. Enzymolysis time, Enzyme amount and enzymolysis temperature have the effect on the extraction of salvianolic acid;the optimum mixture extraction process were as follows: enzymolysis 45 mins,the amount of enzyme was 0.8%,the enzymolysis temperature was 70℃,after that we considered the influence of baptist extracion for salvianolic acid,and the best baptist process were as follows:baptist temperature was 40℃, baptist pH was 4.71,baptist time was 40 min.After optimization the extraction process was stable,reasonable.We prepare Ganoderma lucidum extract by the method,and consider the protective effect Ganoderma extract 1 g·kg-1 can significantly reduce alcohol-induced liver injury in mice ALT, AST activity and malondialdehyde (MDA) content in liver homogenate, to improve their superoxide dismutase (SOD) activity,0.5 g·kg-1 can significantly reduce alcohol-induced liver injury in mice ALT, AST activity,and MDA content in the liver homogenate; and different dose of extract have a good dose-effect relationship. Ganoderma extract powder used in the clinical common oral dose is safe and effective.
