Laser Confocal Scanning Microscopy Research And Software Development

The appearance of LCSM(Laser Confocal Scanning Microscope) is one of the milestones in scientific measurement in 1980s. Because of its ultra resolution and 3D sectioning feature,in no time,LCSM becomes a key tool of the research and application in many areas,such as biomedicine,morphologic,cytogenetics,neurology,pharmacology,etc. Unfortunately,the price of LCSM is so expensive that oneLSM510 costs 2,000,000 RMB ake. In addition,LCSM is forestalled by developedcountries,and is blank for our country. In order to change our embarrassed position radically,our laboratory began to set foot in LCSM at 1998,and the research is progressing rapidly,up to now,the theoretic research and the first-generation LCSM have been performed. In this system,3D visualization is another focus of our research besides the complex hardware so as to satisfy customer's requirements. AMC(Advanced Marching Cubes) algorithm is presented to complete the key step of visualization and the related image software has developed,which named Sunnico Image Management System for Confocal Scanning Microscope. This system has received high evaluation in the High Technology Exhibition in Shenzhen at Oct. 2001. There are six chapters in all in this thesis. The first chapter gives a general description of most of the 3D microscopes nowadays and emphasizes the CSM. In the second chapter,imaging feature of the CSM is analyzed after the description of the architecture of our reflection LCSM. The third chapter describes visualization,including its concept,reference model,usage,classification,method,application,etc. In the fourth chapter,the design and implementation of our image software system is-discussed and the three kernel algorithms are analyzed in detail. The fifth gives a solution of our first generation prototype and the corresponding experimental results. In the last chapter,chapter six,the summary and prospect are given.
