The research studies on the optimize of media and culture condition in the fennentation of Cordyceps Militarism(L.Fr) Link in-depthly and systematically. Under the optimal media and culture condition, the culture was enlarged. Then the paper studies on the examination of the content, extraction and purification technics of polysaccharide in mycelia and in fermentation liquid, cordycepin in mycelia separately. The result is that:1. To do single factor experiment analysis of pitching rate (%), culture temperature (℃) , initial pH and culture time(h). According to the single factor experiment, the optimal technique parameter of fermention was researched by orthogonal design. The best fermention condition is: Pitching rate 10%, culture temperature 27℃, initial pH7, culture time96h.2. Using dry weight of mycelia and weight of crude polysaccharide as the index, all of glucose, malt sugar, refined sugar, sugar, soluble starch showed the significance difference at 0.05 level(P<0.05).Choosing glucose as the best carbon source, the carbon source concentration should control between 4.0-5.0g/L.3. Using dry weight of mycelia and weight of crude polysaccharide as the index, all of Yeast extract, Peptone, Ammonium chloride, Sodium nitrate, beef extract showed the significance difference at 0.05 level(P<0.05). Choosing peptone as the best nitrogen source, the most suitable nitrogen source concentration is 2.0g/L.4. Choosing glucose, peptone, KH2PO4, FeSO4 as raw material, the orthogonal design experiment was taken. The influence of the four factors to the dry weight of mycelia from top to low is: glucose, peptone, KH2PO4 and FeSO4. The best fennentation media should be: glucose 5g/100 ml, peptone 0.2g/100ml, FeSO4 0.001g/100ml, KH2PO4 0.05 g/100ml.5. To do extraction, purification and examination to polysaccharide in mycelia and fementation liquid by sevage and polyglucosan column chromatography, we got refined polysaccharide. The rate is: polysaccharide in dry mycelia is 1.7228g/100g;polysaccharide in fementation liquid is 0.1007g/L.The content of polysaccharide in dry mycelia is 70.30%;the content of polysaccharide in fermentation liquid is 66.9%.6. To do extraction, separation and purification by anion exchange resin and examination by HPLC to cordycepin in dry mycelia, cordycepin crystalline was gotten. The rate is 53.88mg/100g in dry mycelia. The rate of cordycepin in cordycepin crystalline is 88.9%.
